目的:探讨乌鳖口服液对自身免疫性卵巢早衰小鼠的影响。方法:通过皮下多点注射小鼠透明带建立自身免疫性卵巢早衰小鼠模型。然后连续4周给予乌鳖口服液进行治疗,每天剂量为(16.5 g/kg)。通过酶联免疫法检测小鼠抗透明带抗体,用免疫组化法测定脾脏T细胞CD_4~+、CD_8~+的含量。同时用免疫组化法测定卵巢Fas、Fas L蛋白的表达。卵巢和脾脏做病理组织学的分析。结果:模型组小鼠抗透明带抗体的浓度(8.74±0.99)U/m L和CD_4~+/CD_8~+比值(1.74±0.31)增加,同时,模型组小鼠卵巢组织的Fas蛋白表达(0.54±0.29)减少,Fas-L蛋白表达(2.25±0.0)增加。与模型组比较,乌鳖口服液组小鼠抗透明带抗体的浓度(7.34±1.21)U/m L明显低于模型组(P<0.05)。脾脏CD_4~+/CD_8~+比值(1.07±0.14)明显低于模型组(P<0.01)。乌鳖口服液组FasL蛋白表达(1.29±0.86)明显低于模型组(P<0.05)。结论:乌鳖口服液具有降低免疫性卵巢早衰小鼠外周血抗透明带抗体浓度的升高,调节CD_4~+/CD_8~+比值和Fas/FasL系统的作用。
Objective: To investigate the effects of Wu Tui oral liquid on autoimmune premature ovarian failure in mice. Methods: A mouse model of autoimmune premature ovarian failure was established by subcutaneous injection of mouse zona pellucida. Conclusion: Wu Tuai oral liquid can reduce the anti-zona pellucida antibody concentration in peripheral blood of immune-premature ovarian failure and regulate the ratio of CD_4 ~ + / CD_8 ~ + and Fas / FasL system.
