了解雌激素对大鼠胰腺腺泡细胞癌癌前病变及其K-ras基因突变的影响。方法雄性Wistar大鼠单次腹腔注射重氮乙酰丝氨酸(azaserine)30mg/kg体重,部分大鼠2个月后开始每2周内肌肉注射成酸雌二醇0.18mg/100g体重,4个月后实验结束常规取胰腺组织行光镜和电镜检查,并应用PCR-SSCP法检测K-ras基因突变情况。结果单纯给予Azaserine的大鼠胰腺腺泡细胞癌癌前病变(AACN)发生率100%,K-ras基因突变率42.1%;经雌激素处理后,AACN发生率92.4%,K-ras基因突变率25.6%。结论Azaserine可成功诱导大鼠胰腺腺泡细胞癌癌前病变,K-ras基因突变在此过程中属早期事件。雌激素在一定程度上可抑制该癌癌前病变的发生发展。
To understand the effects of estrogen on precancerous lesions of pancreatic acinar cells and K-ras mutations in rats. Methods Male Wistar rats were given a single intraperitoneal injection of aza-ase 30 mg/kg body weight. Some rats started intramuscular injection of 0.18 mg/100 g body weight of estradiol 2 weeks after 2 months. After the end of the experiment, the pancreatic tissue was routinely taken for light microscopy and electron microscopy, and the K-ras gene mutation was detected by PCR-SSCP. Results The incidence of precancerous lesion of pancreatic acinar cell (AACN) was 100% in rats treated with Azaserine alone, and the mutation rate of K-ras was 42.1%. After treatment with estrogen, the incidence of AACN was 92.4%. K- The mutation rate of ras gene is 25.6%. Conclusion Azaserine can successfully induce precancerous lesion of pancreatic acinar cell carcinoma in rats. K-ras gene mutation is an early event in this process. Estrogen can inhibit the development of precancerous lesions to some extent.
