利用荧光探针和免疫组织化学方法,研究了乌贼墨对H22癌细胞内Ca2+浓度、核膜Ca2+/Mg2+-ATP酶活性及c-jun表达的影响。结果发现,乌贼墨使H22癌细胞内Ca2+浓度分别降低了69%和79%,核膜Ca2+/Mg2+-ATP酶活性分别降低了21%和37%,c-jun表达明显减少。结果提示乌贼墨可能通过降低细胞内Ca2+浓度,继而影响到核膜上Ca2+依赖性Ca2+/Mg2+-ATP酶活性,减少了Ca2+向细胞核内的转运,使Ca2+对c-jun表达的促进作用减弱,抑制了细胞的分裂增殖。这可能是乌贼墨抑制肿瘤生长的机制之一。
Using fluorescence probes and immunohistochemical methods, the effects of sepia ink on Ca2+ concentration, nuclear membrane Ca2+/Mg2+-ATPase activity, and c-jun expression in H22 cancer cells were studied. The results showed that squid ink reduced intracellular Ca2+ concentration in H22 cancer cells by 69% and 79%, and nuclear membrane Ca2+/Mg2+-ATPase activity decreased by 21% and 37%, respectively, and c-jun expression was significantly reduced.
