目的 探讨氧化应激对肺泡表面活性物质结合蛋白A(SPA)表达的调控作用。 方法 提取SPA转录因子 (TTF1 ) ,采用凝胶电泳迟滞试验 (EMSA)及核转录试验 ,测定氧化应激对TTF1 与SPA基因调控区DNA的结合活性及SPA核转录活性。 结果 过氧化氢 (H2 O2 )剂量依赖地抑制TTF1 与SPA基因调控区DNA的结合及抑制SPA转录活性。 结论 氧化应激对TTF1 活性的抑制可能是急性肺损伤SPA表达下降的重要原因。
Objective To investigate the regulation of oxidative stress on the expression of alveolar surfactant protein A (SPA). Methods SPA transcription factor (TTF1) was extracted. The DNA binding activity of TTF1 and SPA gene regulatory region and nuclear transcription activity of SPA were determined by EMSA and nuclear transcription assay. Results Hydrogen peroxide (H2O2) dose-dependently inhibited the DNA binding between TTF1 and SPA regulatory region and inhibited SPA transcription activity. Conclusion The inhibition of TTF1 activity by oxidative stress may be the important reason for the decrease of SPA expression in acute lung injury.
