目的探讨注射金丝桃素(hypericin,HY)并525nm激光照射对鲜红斑痣模型罗曼鸡鸡冠的作用。方法取6月龄雄性罗曼鸡36只,按随机数字表法分为6组:正常对照组、激光对照组、血卟啉1mg/kg阳性对照组、HY0.3mg/kg组、HY1mg/kg组、HY3mg/kg组,每组6只。除正常对照组外,对各组鸡冠单侧进行激光照射,未处理一侧作为自身对照。罗曼鸡静脉注射金丝桃素后,立即用激光照射鸡冠直径2cm的区域,鸡冠剂量为功率密度100mW/cm2。各组(除正常对照组)给药并525nm激光照射后每天观察照相,处理7、14d后处死动物,取鸡冠进行病理检查,并对鸡冠毛细血管计数。结果HY给药组和血卟啉对照组从外观上可对鸡冠表皮产生不同程度的损伤,造成明显水肿、结痂、坏死。HY1mg/kg,HY3mg/kg组及血卟啉1mg/kg阳性对照组对鸡冠的表皮损伤尤其明显,产生明显的坏死、结痂。HE组织学观察显示,HY各给药组及血卟啉对照组能导致鸡冠表皮增生,减少真皮毛细血管数,其毛细血管减少率与正常对照组及激光对照组相比差异有统计学意义(P<0.01),激光对照组与正常对照组相比差异不大。结论HY静脉注射给药并525nm激光照射对鸡冠真皮毛细血管有较好的破坏作用,能对鲜红斑痣模型鸡冠起到明显损伤作用。
Objective To investigate the effect of hypericin (HY) injection and 525nm laser irradiation on Portland roman chicken cockscomb model. Methods Sixty-six male Roman broilers aged 6 months were randomly divided into 6 groups according to random number table: normal control group, laser control group, hematoporphyrin 1mg / kg positive control group, HY0.3mg / kg group, HY1mg / kg group , HY3mg / kg group, 6 in each group. In addition to the normal control group, the unilateral treatment of each group was laser irradiation, untreated side as a self-control. Romans chicken intravenous injection of hypericin immediately after the laser irradiation of the corona crown diameter 2cm area, the crest dose power density 100mW / cm2. The rats in each group (except the normal control group) were dosed daily and photographed after 525nm laser irradiation. The animals were sacrificed on the 7th and 14th day. The animals were sacrificed and the canopy was taken for pathological examination. The capillaries were counted. Results The HY group and the hematoporphyrin group showed different degrees of damage to the epidermis of cockscomb in appearance, resulting in obvious edema, crusting and necrosis. HY1mg / kg, HY3mg / kg group and hematoporphyrin 1mg / kg positive control group of chicken crown epidermal injury is particularly evident, resulting in significant necrosis, crusting. Histological observation showed that HY administration group and hematoporphyrin control group could lead to epidermal hyperplasia and reduce the number of dermal capillaries, the capillary reduction rate was significantly different from the normal control group and the laser control group ( P <0.01). There was no significant difference between the laser control group and the normal control group. Conclusion HY intravenous injection and 525nm laser irradiation have a good destructive effect on the dermal capillaries of the cocks, which can obviously damage the crest of the port wine stain model.
