目的:探讨牛奶对金黄地鼠皮脂分泌的影响及作用机制。方法:将18只金黄地鼠随机分为3组,每组6只。空白组不予任何干预措施,全脂牛奶组予全脂牛奶灌胃,脱脂牛奶组予脱脂牛奶灌胃,2.5 ml/次,2次/d,连续4周;于干预开始后第0、7、14、21、28天测量金黄地鼠双侧皮脂腺斑最大横径及纵径,计算皮脂腺斑的面积;末次灌胃24 h后剪下各组金黄地鼠双侧皮脂腺斑组织,采用免疫组化法检测皮脂腺斑胰岛素样生长因子1(IGF-1)/固醇调节元件结合蛋白-1(SREBP-1)/乙酰辅酶A羧化酶(ACC-1)信号通路表达水平。采用重复测量方差分析、成组设计的单因素方差分析、Kruskal-Wallis n H检验进行统计学分析,两两比较采用LSD-n t检验。n 结果:重复测量方差分析显示,金黄地鼠各组间皮脂腺斑面积差异无统计学意义(n F= 0.96,n P= 0.417)。脱脂牛奶组IGF-1表达(0.39 ± 0.03)显著高于空白组(0.35 ± 0.03)及全脂牛奶组(0.33 ± 0.02),差异均有统计学意义(n t值分别为2.62、3.82,n P值分别为0.021、0.002);与空白组(0.36 ± 0.02)相比,脱脂牛奶组SREBP-1表达显著升高(0.42 ± 0.04,n t= 2.64,n P= 0.021);与空白组ACC-1表达(0.34 ± 0.03)相比,全脂牛奶组(0.40 ± 0.03)及脱脂牛奶组(0.40 ± 0.05)均显著升高(n t值分别为2.39、2.47,n P值分别为0.031、0.026)。n 结论:牛奶可能通过IGF-1/SREBP-1/ACC-1信号通路促进金黄地鼠皮脂分泌。“,”Objective:To investigate the effect of milk on sebum secretion in golden hamsters, and to explore its possible mechanism of action.Methods:Eighteen golden hamsters were randomly and equally divided into 3 groups: blank control group receiving no intervention, whole-milk group gavaged with whole milk, and skimmed-milk group gavaged with skimmed milk. The gavage feeding was performed at a dose of 2.5 ml twice a day for 4 consecutive weeks. The maximum transverse diameter and maximum longitudinal diameter of bilateral sebaceous gland spots were measured on days 0, 7, 14, 21 and 28 after the start of intervention, and the area of sebaceous gland spots was calculated; at 24 hours after the last gavage, bilateral sebaceous gland spot tissues were resected, and subjected to immunohistochemical study to determine the expression of insulin-like growth factor-1 (IGF-1) /sterol regulatory element-binding protein-1 (SREBP-1) /acetyl-coenzyme A carboxylase (ACC-1) signaling pathway in sebaceous gland spots. Statistical analysis was carried out by using repeated measures analysis of variance, one-way analysis of variance for independent groups, Kruskal-Wallis n H test, and least significant difference-n t test for multiple comparisons.n Results:Repeated measures analysis of variance showed that there was no significant difference in the area of sebaceous gland spots of golden hamsters among the 3 groups (n F= 0.96, n P= 0.417) . The IGF-1 expression was significantly higher in the skimmed-milk group (0.39 ± 0.03) than in the blank control group (0.35 ± 0.03, n t= 2.62, n P= 0.021) and whole-milk group (0.33 ± 0.02, n t= 3.82, n P= 0.002) ; compared with the blank control group (0.36 ± 0.02) , the skimmed-milk group showed significantly increased SREBP-1 expression (0.42 ± 0.04, n t= 2.64, n P= 0.021) ; the ACC-1 expression was significantly higher in the skimmed-milk group (0.40 ± 0.03) and whole-milk group (0.40 ± 0.05) than in the blank control group (0.34 ± 0.03; n t= 2.39, 2.47, n P= 0.031, 0.026, respectively) .n Conclusion:Milk may promote sebum secretion in golden hamsters through the IGF-1/SREBP-1/ACC-1 signaling pathway.
