目的研究叶酸、维生素B_6(VB_6)和 B_(12)(VB_(12))对局灶性脑缺血大鼠血浆同型半胱氨酸(homocysteine,Hey)水平和学习记忆能力的影响。方法 SD 大鼠按体重随机分为假手术组(ShamOP)、大脑中动脉闭塞模型组(MCAO)、大脑中动脉闭塞+叶酸组(MCAO+FA)和 MCAO+复合维生素(叶酸、VB_6和 VB_(12))组(MCAO+CV),补充前、后和缺血后分别检测大鼠血浆 Hcy 浓度,缺血后进行神经功能评分,测定学习记忆能力。结果大鼠脑缺血后血浆 Hcy 浓度 MCAO+FA 组(6.92±1.04) μmol/L 和 MCAO+CV 组(5.49±1.00)μmol/L 低于 Sham OP 组(9.33±1_11)μmol/L 和MCAO 组(10.90±2.03)μmol/L(P<0.05),且 MCAO+CV 组低于 MCAO+FA 组(P<0.05);神经功能缺失评分 MCAO+FA 组(1.75±0.46)和 MCAO+CV 组(1.38±0.52)以及 Y 型迷宫中受电击次数MCAO+FA 组(123.50±39.77)和 MCAO+CV 组(86.25±21.39)低于 MCAO 组神经功能缺失评分(2.62±0.52)和电击次数(173.25±46.32)(P<0.05),而记忆正确次数 MCAO+CV 组(3.75±0.42)高于 MCAO 组(2.12±0.45)(P<0.05)。结论叶酸能降低脑缺血大鼠血浆 Hcy 浓度,增强脑缺血大鼠学习记忆能力,改善神经功能,且联合补充 VB_6和 VB_(12)优于单独补充叶酸。
Objective To study the effects of folic acid, vitamin B_6 (VB_6) and B_ (12) (VB_ (12)) on plasma homocysteine (Hey) and learning and memory ability in rats with focal cerebral ischemia. Methods SD rats were randomly divided into sham operation group, middle cerebral artery occlusion model group (MCAO), middle cerebral artery occlusion + folate group (MCAO + FA) and MCAO + multivitamin (folic acid, VB_6 and VB 12 ) Group (MCAO + CV). Plasma Hcy levels were measured before, during and after ischemia, and neurological scores were measured after ischemia. Learning and memory ability was measured. Results Compared with Sham OP group (9.33 ± 1_11) μmol / L and MCAO + FA group, the concentrations of Hcy in MCAO + FA group (6.92 ± 1.04) μmol / L and MCAO + CV group (10.90 ± 2.03) μmol / L (P <0.05), and MCAO + CV group was lower than MCAO + FA group (P <0.05); neurological deficit score was significantly higher in MCAO + FA group (1.75 ± 0.46) and MCAO + CV group (1.38 ± 0.52) and the number of electroshock in the Y-type maze were lower than those in MCAO + FA group (123.50 ± 39.77) and MCAO + CV group (86.25 ± 21.39) ± 46.32) (P <0.05), while MCAO + CV group (3.75 ± 0.42) was significantly higher than MCAO group (2.12 ± 0.45) (P <0.05). Conclusion Folic acid can decrease plasma Hcy concentration in cerebral ischemia rats, enhance learning and memory ability and improve neurological function in cerebral ischemic rats. Folic acid supplementation with VB_6 and VB_ (12) is superior to folic acid supplementation alone.
