目的:通过检测瘦素(Leptin)-神经肽Y(NPY)/血管活性肠肽(VIP)/胃泌素(GAS)信号传导通路及胸腺指数、脾指数的变化,探讨不同浓度黄芪建中汤对脾气虚证大鼠的影响。方法:复制脾气虚证大鼠模型(大黄泻下、游泳力竭和饥饱失常三因素复合法),造模大鼠随机分为正常对照组,模型组,四君子汤组,黄芪建中汤低、中、高剂量组,在用药21 d后检测黄芪建中汤对大鼠血清学及组织学Leptin、NPY、VIP、GAS蛋白表达及胸腺指数、脾指数的影响。结果:与模型组比较,黄芪建中汤中、高剂量组血清木糖浓度、CPK、GAS、NPY含量上升,Leptin及VIP含量下降,与模型组比较差异均有统计学意义(P<0.01或P<0.05),与对照组比较差异有统计学意义(P<0.01或P<0.05),尤以中剂量组作用最为明显。在造模成功后,大鼠组织学Leptin、VIP表达增加,GAS、NPY表达降低,同时伴随指数、胸腺指数的降低(P<0.01)。在给予黄芪建中汤治疗21 d后,黄芪建中汤中、高剂量组Leptin、VIP表达降低,GAS、NPY表达增加,胸腺指数、脾指数也相应回升,模型组比较,差异有显著统计学意义(P<0.01),且中剂量组具有最佳的剂量-效应比(P<0.05或P<0.01)。结论:经方黄芪建中汤治疗脾气虚证的机制可能是通过LeptinNPY/VIP/GAS信号转导通路及提高胸腺指数、脾指数发挥作用,且具有明显的剂量效应关系。
OBJECTIVE: To detect the changes of Leptin, NPY / VIP / GAS signal pathways, thymus index and spleen index, to investigate the effects of different concentrations of Astragalus Jianzhong Tang Effect on Spleen Deficiency Syndrome Rats. Methods: The rat model of spleen-qi deficiency syndrome (rhubarb and diarrhea, swimming exhaustion and starvation insufficiency three-factor complex method) was established. The rats were randomly divided into normal control group, model group, Sijunzi Decoction group, , Middle-dose and high-dose groups. The effects of Astragalus Jianzhong Decoction on serum and histological Leptin, NPY, VIP, GAS protein and thymus index and spleen index were detected 21 days after treatment. Results: Compared with the model group, the concentrations of serum xylose, CPK, GAS, NPY and the contents of Leptin and VIP in astragaluschabu medium and high dose groups were significantly lower than those in the model group (P <0.01 or P <0.05). The difference was statistically significant (P <0.01 or P <0.05) compared with the control group, especially in the middle dose group. After successful modeling, the expression of Leptin and VIP increased, the expression of GAS and NPY decreased, accompanied by the decrease of index and thymus index (P <0.01). After given Astragalus Jianzhong Decoction for 21 days, the expression of Leptin and VIP, the expression of GAS and NPY increased, the thymus index and spleen index also increased in middle and high dose of Astragalus Jianzhong Decoction, the difference was statistically significant (P <0.01), and the middle dose group had the best dose-effect ratio (P <0.05 or P <0.01). Conclusion: The mechanism of the prescription Huangqi Jianzhong decoction in the treatment of spleen deficiency syndrome may be through the LeptinNPY / VIP / GAS signal transduction pathway and increase the thymus index, spleen index play a role, and has a significant dose-response relationship.
