目的分析鸟氨酸脱羧酶抗酶抑制子基因在小鼠生长发育过程中的功能。方法用特殊的捕获载体导入小鼠ES细胞中,5′RACE方法鉴定成功地捕获鸟氨酸脱羧酶抗酶抑制子基因(Oazin)后,由这种ES制作了Oazin“敲出”小鼠。结果Oazin“敲出”小鼠捕获载体位于Oazin基因启动子上游,Qazin基冈的转录被抑制。结论Oazin“敲出”小鼠为分析Oazin基因的功能提供了有用的实验材料。
OBJECTIVE: To analyze the function of ornithine decarboxylase antizyme inhibitor gene in mouse growth and development. Methods Mouse ES cells were transfected with a special capture vector and Oazin “knockout” mice were produced from these ESs after successful identification of the ornithine decarboxylase inhibitor (Oazin) by the 5 ’RACE method. Results The Oazin “knockout” mouse capture vector was located upstream of the Oazin gene promoter and the transcription of Qazin was inhibited. Conclusion Oazin “knockout” mice provide useful experimental data for analyzing the function of the Oazin gene.
