目的:探讨丁酸钠对人食管癌Eca-109细胞凋亡的作用。方法:以1 mmol/L、2.5 mmol/L和5 mmol/L丁酸钠分别与人食管癌Eca-109细胞共培养,倒置显微镜观察细胞生长情况,免疫组化(SP)法检测凋亡抑制基因bax蛋白的表达。结果:Eca-109细胞经丁酸钠处理后,在倒置显微镜下观察到丁酸钠处理后的Eca-109细胞形态学发生了改变,细胞生长明显受到抑制;免疫组化检测实验组细胞的bax蛋白表达阳性率低于对照组,具有统计学意义(P<0.05)。结论:丁酸钠可诱导食管癌Eca-109细胞凋亡,其机制与下调凋亡抑制基因bax蛋白表达有关。
Objective: To investigate the effect of sodium butyrate on the apoptosis of human esophageal cancer Eca-109 cells. Methods: The human esophageal cancer cell line Eca-109 was co-cultured with 1 mmol / L, 2.5 mmol / L and 5 mmol / L sodium butyrate respectively. The growth of the cells was observed under inverted microscope and the apoptosis was detected by immunohistochemistry Gene bax protein expression. Results: The morphological changes of Eca-109 cells treated with sodium butyrate were observed under inverted microscope after Eca-109 cells were treated with sodium butyrate. The growth of Eca-109 cells was obviously inhibited. The expression of bax The positive rate of protein expression was lower than the control group, with statistical significance (P <0.05). CONCLUSION: Sodium butyrate can induce the apoptosis of esophageal cancer Eca-109 cells. The mechanism is related to the down-regulation of bax protein.
