目的探讨肿瘤坏死因子受体相关因子-6(tumor necrosis factor receptor-associated factor-6,TRAF-6)在炎症性肠病患者外周血单个核细胞和血浆的表达,及其与内镜下疾病活动性的相关性。方法应用酶联免疫吸附试验(ELISA)分析炎症性肠病患者和正常对照者血浆中TRAF-6蛋白的表达,SYBR-green real time PCR方法分析炎症性肠病患者和正常对照者外周血单个核细胞中TRAF-6mRNA的表达。数据处理使用Graph Pad Prism5。结果克罗恩病患者(P=0.000)和溃疡性结肠炎患者(P=0.000)血浆TRAF-6水平显著高于正常对照者,但是克罗恩病患者(r=-0.114,P=0.377)以及溃疡性结肠炎患者(r=0.044,P=0.727)血浆TRAF-6表达水平和内镜下疾病活动指数均无显著的相关性。克罗恩病患者(P=0.000)和溃疡性结肠炎(P=0.000)患者外周血单个核细胞TRAF-6mRNA表达均显著高于正常对照者外周血单个核细胞中mRNA的表达。结论炎症性肠病患者血浆和外周血单个核细胞中均存在TRAF-6的激活,但血浆TRAF-6的高表达不能反映内镜下的疾病活动程度。
Objective To investigate the expression of tumor necrosis factor receptor-associated factor-6 (TRAF-6) in peripheral blood mononuclear cells and plasma in patients with inflammatory bowel disease and its relationship with endoscopic disease activity Sexual relevance. Methods The expression of TRAF-6 protein in plasma of patients with inflammatory bowel disease and normal controls was analyzed by enzyme linked immunosorbent assay (ELISA). SYBR-green real time PCR method was used to analyze the expression of TRAF-6 in peripheral blood of patients with inflammatory bowel disease and normal controls TRAF-6 mRNA expression in cells. Data processing uses Graph Pad Prism5. Results Plasma levels of TRAF-6 in Crohn’s disease patients (P = 0.000) and ulcerative colitis patients (P = 0.000) were significantly higher than those in normal controls, but patients with Crohn’s disease (r = -0.114, P = 0.377) (R = 0.044, P = 0.727). There was no significant correlation between the level of TRAF-6 in plasma and the activity index of disease under endoscopy in patients with ulcerative colitis. The expression of TRAF-6 mRNA in peripheral blood mononuclear cells in patients with Crohn’s disease (P = 0.000) and ulcerative colitis (P = 0.000) was significantly higher than that in normal controls. Conclusion The activation of TRAF-6 exists in both plasma and peripheral blood mononuclear cells in patients with inflammatory bowel disease. However, the high expression of TRAF-6 in plasma does not reflect the degree of endoscopic activity.
