目的探讨CXXC4对卵巢癌细胞的增殖及药物敏感性的影响。方法采用Western blotting和Real-time PCR分别从蛋白水平和mRNA水平检测正常卵巢上皮细胞和卵巢癌细胞中的CXXC4表达,脂质体法将重组质粒pc DNA3-CXXC4和siRNA-CXXC4分别转染人卵巢癌细胞SKOV3、Caov3和正常卵巢上皮细胞IOSE80,CCK8法分析CXXC4对细胞增殖及药物敏感性的影响。结果与正常卵巢上皮细胞相比,卵巢癌细胞中CXXC4低表达,外源性CXXC4过表达抑制卵巢癌细胞SKOV3、Caov3的增殖能力,以及增强肿瘤细胞对化疗药物的敏感性;降低CXXC4的表达能促进正常卵巢上皮细胞的增殖能力,促进细胞对化疗药物的耐受。结论 CXXC4的过表达抑制卵巢癌细胞的增殖能力,且增强肿瘤细胞对化疗药物的敏感性,提示其可能作为一个潜在的卵巢癌疗效预测标记物以及化疗增敏的分子靶点。
Objective To investigate the effect of CXXC4 on the proliferation and drug sensitivity of ovarian cancer cells. Methods The expression of CXXC4 in normal ovarian epithelial cells and ovarian cancer cells was detected by Western blotting and Real-time PCR respectively. The recombinant plasmid pcDNA3-CXXC4 and siRNA-CXXC4 were transfected into human ovary The effect of CXXC4 on proliferation and drug sensitivity of cancer cells SKOV3, Caov3 and normal ovarian epithelial cells IOSE80 and CCK8 were analyzed. Results Compared with normal ovarian epithelial cells, the expression of CXXC4 in ovarian cancer cells was low, and the exogenous CXXC4 overexpression inhibited the proliferation of ovarian cancer cells SKOV3 and Caov3 as well as the sensitivity of tumor cells to chemotherapeutic drugs. The expression of CXXC4 decreased Promote normal ovarian epithelial cell proliferation, promote cell tolerance to chemotherapy drugs. Conclusion The overexpression of CXXC4 inhibits the proliferation of ovarian cancer cells and enhances the sensitivity of tumor cells to chemotherapeutic drugs, suggesting that CXXC4 may serve as a potential molecular marker for predicting ovarian cancer efficacy and chemosensitivity.
