水稻细菌性条斑病菌(Xamthomonas.oryzae pv.oryzicola,Xoc)是水稻的主要病原菌之一,其引发的水稻细菌性条斑病可造成水稻严重减产。本文利用Xoc GX01作为实验菌株,通过细菌培养、蛋白提取、纯化获得总蛋白,利用高p H值C18反相色谱作为第一维分离方式,再将收集的馏分通过LC-MS/MS技术得到了总蛋白质表达谱,共鉴定蛋白质1 621个,同时利用生物信息学技术对鉴定蛋白进行基因本体(gene ontology,GO)生物学分类,对生物功能、生物进程以及细胞组件进行了注释。为明确植物病原菌的作用机理和研究植物-病原微生物互作提供了方法学基础。
Xam is one of the major pathogenic bacteria in rice, and the rice bacterial leaf streak caused by it can cause serious rice yield loss. In this paper, Xoc GX01 was used as the experimental strain to obtain the total protein by bacterial culture, protein extraction and purification. The high p H value C18 reversed phase chromatography was used as the first dimension separation method. The collected fractions were obtained by LC-MS / MS A total of 1621 proteins were identified. At the same time, bioinformatics techniques were used to classify the identified proteins by gene ontology (GO). The biological functions, biological processes and cell components were annotated. It provides a methodological basis for clarifying the mechanism of action of plant pathogens and studying the interaction of plant-pathogenic microorganisms.
