目的测定从块菌菌丝体中分离得到的均一多糖KJB-1的平均相对分子质量,并分析其单糖组成。方法块菌菌丝体通过水提醇沉,再以Sevage法去除蛋白质,得到粗多糖KJB。经Sepharose CL6B柱色谱分离纯化得到均一多糖KJB-1。利用高效液相色谱法测定KJB-1的平均相对分子质量;三氟乙酸完全酸水解后进行糖醇衍生化,之后运用气相色谱法,与标准品对照后分析其单糖组成。结果KJB-1在高效液相下呈单一对称峰,其平均相对分子质量大于2×106,单糖组成分析该多糖是由单一组分葡萄糖组成。结论KJB-1为分子质量大于2×106的葡聚糖。
Objective To determine the average relative molecular mass of homogeneous polysaccharide KJB-1 isolated from truffle mycelium and to analyze its monosaccharide composition. Methods The truffle mycelia were extracted by alcohol and then the proteins were removed by Sevage method to obtain crude polysaccharide KJB. Purification by Sepharose CL6B column chromatography to obtain homogeneous polysaccharide KJB-1. The average relative molecular mass of KJB-1 was determined by HPLC. Trifluoroacetic acid was completely acid-hydrolyzed and then derivatized with alditol. The monosaccharide composition of the KJB-1 was analyzed by gas chromatography. Results KJB-1 showed a single symmetrical peak under high-performance liquid phase. The average molecular weight of KJB-1 was more than 2 × 106. Monosaccharide composition analysis The polysaccharide consisted of a single component of glucose. Conclusion KJB-1 is a dextran with molecular weight greater than 2 × 106.
