目的探讨microRNA-206(miRNA-206)单核苷酸多态性与乙型肝炎病毒(HBV)相关原发性肝癌遗传易感性及术后复发的关系。方法共纳入三组受试对象,分别为128例HBV相关肝癌患者(HCC组)、121例慢乙肝患者(CHB组)及129例健康受试者(CON组)。采用生物信息学方法预测并筛选位于miRNA-206启动子调控区域的三个标签SNP,基质辅助激光解吸电离飞行时间质谱法检测miRNA-206 rs16882131、rs6920648、rs945210三个SNP位点的基因型,分析其与HBV相关肝癌易感性及早期复发的关系。结果 miRNA-206 rs16882131基因型及等位基因分布在HCC、CHB、CON三组间差异有统计学意义,其突变基因型CC和等位基因C降低HBV相关肝癌发生风险(OR=0.354,95%CI0.163~0.771;OR=0.182,95%CI 0.124~0.267),但与肝癌术后早期复发无显著相关性。结论 miRNA-206 rs16882131多态性可能与HBV相关肝癌遗传易感性相关,但与肝癌术后早期复发无明显关系。
Objective To investigate the relationship between microRNA-206 (miRNA-206) single nucleotide polymorphisms and genetic predisposition and postoperative recurrence of hepatitis B virus (HBV) -related primary hepatocellular carcinoma (HCC). Methods A total of three groups of subjects were enrolled: 128 HBV-related hepatocellular carcinoma patients (HCC group), 121 chronic hepatitis B patients (CHB group) and 129 healthy subjects (CON group). The bioinformatics methods were used to predict and screen the three SNPs located in the regulation region of miRNA-206 promoter. The genotypes of miRNA-206 rs16882131, rs6920648 and rs945210 were detected by matrix-assisted laser desorption / ionization time-of-flight mass spectrometry Its relationship with HBV-related susceptibility and early recurrence of liver cancer. Results The genotype and allele distribution of miRNA-206 rs16882131 in HCC, CHB and CON groups were statistically significant. The genotype CC and allele C of mutant miRNA-206 reduced the risk of HBV-related HCC (OR = 0.354, 95% CI0.163 ~ 0.771; OR = 0.182, 95% CI 0.124 ~ 0.267), but no significant correlation with early postoperative recurrence of liver cancer. Conclusion The polymorphism of miRNA-206 rs16882131 may be related to the genetic susceptibility to HBV-related hepatocellular carcinoma, but not to the early recurrence of hepatocellular carcinoma.
