目的观察高脂饮食对C57BL/6小鼠出现外周神经病变的影响。方法将3周龄雌性C57BL/6小鼠随机分为正常对照(NC)组和高脂饮食(HF)组,隔周行腹腔内葡萄糖耐量实验,于HF组小鼠开始出现糖耐量异常时行热痛觉及神经传导速度测定,并检测血清中脂质水平、总超氧化物歧化酶(T-SOD)活性和丙二醛(MDA)含量。结果分组喂养14周后,HF组小鼠出现糖耐量异常,热反应时间缩短,运动和感觉神经传导速度均明显减慢(P均<0.05)。TC、HDL-C、LDL-C分别较NC组升高86.99%、38.0%、85.42%(P均<0.01),血清中T-SOD活性及MDA含量均显著升高(P均<0.05)。结论高脂饮食可诱导C57BL/6小鼠发生外周神经病变,血脂紊乱及氧化应激可能是危险因素。
Objective To observe the effect of high-fat diet on peripheral neuropathy in C57BL / 6 mice. Methods Three-week-old female C57BL / 6 mice were randomly divided into normal control (NC) group and high fat diet (HF) group, intraperitoneal glucose tolerance test was performed every other week. When HF group mice began to have impaired glucose tolerance Thermal pain and nerve conduction velocity were measured. The levels of serum lipids, total superoxide dismutase (T-SOD) and malondialdehyde (MDA) were measured. Results After fed for 14 weeks, the mice in HF group had impaired glucose tolerance, shortened the thermal response time and decreased the motor and sensory nerve conduction velocity (all P <0.05). TC, HDL-C and LDL-C were increased by 86.99%, 38.0% and 85.42% respectively in NC group (P <0.01), and the serum T-SOD activity and MDA content were significantly increased (all P <0.05). Conclusion High-fat diet can induce peripheral neuropathy, dyslipidemia and oxidative stress in C57BL / 6 mice may be risk factors.
