目的研究太白楤木抗肝损伤作用活性部位HPLC指纹图谱。方法采用高效液相色谱法,Accurasil-C18柱(250 mm×4.6 mm,5μm),乙腈-0.1%磷酸水梯度洗脱,0~10 min A由5%升至20%,10~25 min A由20%升至28%,25~35 min A由28%升至33%,35~45 min A由33%升至35%,45~55 min A由35%升至50%,55~60 min A由50%升至60%,60~70 minA由60%升至95%,70~80 minA由95%降至5%;检测波长为203 nm;柱温:30℃;流速:0.8 mL/min。采用《指纹图谱相似度评价系统2004A版》对结果进行评价分析。结果确定了太白楤木抗肝损伤作用活性部位指纹图谱共有模式及17个共有峰。10批样品与对照指纹图谱相比,相似度平均值在0.9以上。结论此方法所建立的对照指纹图谱具有较好的代表性,能够用于太白楤木抗肝损伤活性部位的指纹图谱测定。
OBJECTIVE To study the HPLC fingerprinting of the active site against the liver injury of Taibai alder. Methods High performance liquid chromatography (HPLC) was performed on an Accurasil-C18 column (250 mm × 4.6 mm, 5 μm) with a gradient of acetonitrile to 0.1% phosphoric acid. The concentration of A was increased from 5% to 20% From 20% to 28%, A increased from 28% to 33% at 25 to 35 minutes, from 33% to 35% at 35 to 45 minutes, from 35% to 50% at 45 to 55 minutes, and from 55 to 60 min A increased from 50% to 60%, 60-70 min increased from 60% to 95%, 70-80 min decreased from 95% to 5%, the detection wavelength was 203 nm, the column temperature was 30 ℃, the flow rate was 0.8 mL / min. Using “fingerprint similarity evaluation system 2004A version” to evaluate the results of the analysis. Results The common pattern and 17 common peaks of the active site fingerprints of the anti-hepatic injury were identified. Ten batches of samples compared with the control fingerprints, the average similarity of 0.9 or more. Conclusion The control fingerprinting established by this method has good representativeness and can be used for the determination of the anti-hepatic injury active site of Taibai alnum.
