目的:探讨寡糖sLeX与L-选择素(L-selectin)作为黏附分子在母-胎之间的识别与黏附的作用。方法:通过免疫荧光方法检测胚胎细胞(JAR细胞)与子宫内膜细胞(RL95-2细胞)的sLeX与L-选择素的表达,通过转染α1,3-岩藻糖基转移酶VII(fucosyltransferase VII,FUT7)过表达载体至JAR细胞和RL95-2细胞,调节细胞表面sLeX的表达,以及通过sLeX/L-选择素抗体封闭细胞,计算不同处理后JAR细胞与RL95-2细胞黏附率。结果:①胚胎细胞与子宫内膜细胞均表达sLeX和L-选择素;②sLeX抗体及L-选择素抗体分别封闭JAR细胞与RL95-2细胞后,JAR细胞与RL95-2细胞的黏附率下降;③FUT7过表达增加sLeX寡糖抗原的合成,促进JAR细胞与RL95-2细胞的黏附率。结论:sLeX/L-选择素组成的黏附系统在胚胎与子宫内膜的识别与黏附过程中发挥重要作用。
OBJECTIVE: To investigate the role of oligosaccharides sLeX and L-selectin as adhesion molecules in maternal-fetal recognition and adhesion. Methods: The expression of sLeX and L-selectin in embryonic cells (JAR cells) and endometrial cells (RL95-2 cells) was detected by immunofluorescence method. The expression of sLeX and L-selectin was detected by transfecting α1,3-fucosyltransferase VII, FUT7) overexpression vector to JAR cells and RL95-2 cells to regulate the expression of sLeX on the cell surface, and the cells were blocked by sLeX / L-selectin antibody to calculate the adhesion rate between JAR cells and RL95-2 cells. Results: ① Both sLeX and L-selectin were expressed in embryonic cells and endometrial cells; ② The adhesion rate of JAR cells to RL95-2 cells was decreased after sLeX antibody and L-selectin antibody blocked JAR cells and RL95-2 cells respectively; ③FUT7 overexpression increased sLeX oligosaccharide antigen synthesis, and promote JAR cells and RL95-2 cell adhesion rate. Conclusion: The sLeX / L-selectin adhesion system plays an important role in the recognition and adhesion of embryos and endometrium.
