[目的]研究双酚A(bisphenol A,BPA)经口亚急性染毒对雌性小鼠卵巢结构和功能的影响,探讨BPA是否有可能引起卵巢早衰。[方法]5周龄SPF清洁级昆明雌性小鼠经检疫1周后,将小鼠随机分为溶剂对照组和低、中、高剂量组,每组10只小鼠。低、中、高剂量组的染毒剂量分别为40、120、240mg/kg,配成相应的玉米油溶液,按0.4%体重灌胃,对照组用等体积玉米油灌胃。染毒25d后观察其组织病理学,用酶联免疫吸附法(ELISA)测量血清雌二醇(E2)、小鼠抗卵巢抗体(AOAb)水平,用免疫组化方法观察bax、bcl-2表达情况。[结果]与对照组相比,低、中、高染毒剂量组的小鼠E2浓度升高(P<0.05),分别为(13.79±2.36)、(19.68±1.34)、(22.35±1.26)pg/mL;AOAb浓度亦升高(P<0.05),分别为(3.11±0.24)、(3.99±0.56)、(4.51±0.25)ng/mL;bax表达增加,bcl-2表达减少。[结论]BPA可引起小鼠卵巢功能与结构明显变化以及自身免疫反应,但尚不能证明其引起昆明小鼠卵巢早衰。
[Objective] To investigate the influence of subacute exposure to bisphenol A (BPA) on the ovarian structure and function in female mice and explore whether BPA may cause premature ovarian failure. [Method] Five weeks old female Kunming SPF mice were quarantined for one week. The mice were randomly divided into solvent control group and low, medium and high dose groups, 10 mice in each group. The doses of low, medium and high dose groups were 40,120,240 mg / kg, respectively, and the corresponding corn oil solution was dosed by 0.4% body weight. The control group was fed with the same volume of corn oil. Twenty-five days after exposure, the histopathology was observed. Serum estradiol (E2) and anti-ovarian antibodies (AOAb) were measured by enzyme-linked immunosorbent assay (ELISA). The expressions of bax and bcl- Happening. [Results] Compared with the control group, E2 concentrations in the low, middle and high dose groups were significantly higher than those in the control group (P <0.05), which were (13.79 ± 2.36), (19.68 ± 1.34) and (22.35 ± 1.26) pg / mL and AOAb concentrations (3.11 ± 0.24, 3.99 ± 0.56 and 4.51 ± 0.25 ng / mL, respectively). The expression of bax increased and the expression of bcl-2 decreased. [Conclusion] BPA can cause obvious changes of ovarian function and structure and autoimmunity in mice, but it can not prove that it causes premature ovarian failure in Kunming mice.
