目的探讨N-乙酰半胱氨酸(NAC)对糖尿病大鼠心肌肥大的防治作用及机制。方法雄性SD大鼠随机分为正常对照(NC)组、正常对照治疗(NC+NAC)组、糖尿病(DM)组、糖尿病治疗(DM+NAC)组。NC+NAC和DM+NAC组给予NAC 1.4~1.5g·kg~(-1)·d~(-1),持续8周后检测各组心脏收缩压、心率、左室内压舒张速率(-dp/dt)及到达最大舒张速率的时间(T值)、PG、血清Ins、15-F_(2t)-异前列烷(15 F_(2t)-Isop)和超氧化物歧化酶(SO)D)水平。处死动物后测量心肌细胞面积并计算左室质量指数(LVW/BW)。结果 (1)DM+NAC组LVW/BW和心肌细胞面积显著低于DM组(P<0.05),而-dp/dt显著增加,T值显著减少;(2)与DM组比较,DM+NAC组15-F_(2t)-Isop水平显著下降,SOD水平显著升高,PG明显下降(P均<0.05)。结论 NAC可有效抑制糖尿病大鼠心肌肥大,改善心脏舒张功能,这与其增强心肌抗氧化能力有关。
Objective To investigate the preventive and therapeutic effects of N-acetylcysteine (NAC) on cardiac hypertrophy in diabetic rats and its mechanism. Methods Male Sprague-Dawley rats were randomly divided into normal control group (NC), normal control group (NC + NAC), diabetes mellitus (DM) group and diabetes mellitus (DM + NAC) group. NAC was administrated in NC + NAC and DM + NAC groups for 1.4 ~ 1.5 g · kg -1 d -1 for 8 weeks. The systolic pressure, heart rate and left ventricular diastolic velocity (-dp / dt) and the time to maximum diastolic velocity (T value), PG, serum Ins, 15-F 2t -isoprostane and superoxide dismutase (SO D) Level. Cardiac myocyte area was measured after the animals were sacrificed and the left ventricular mass index (LVW / BW) was calculated. Results (1) The LVW / BW and the area of myocardial cells in DM + NAC group were significantly lower than those in DM group (P <0.05), but the level of -dp / dt was significantly increased The level of 15-F_ (2t) -Isop was significantly decreased, the level of SOD was significantly increased and the level of PG was significantly decreased (all P <0.05). Conclusion NAC can effectively inhibit cardiac hypertrophy and improve diastolic function in diabetic rats, which is related to its ability to enhance myocardial antioxidant capacity.
