目的通过检测凋亡抑制基因bc1- 2蛋白和突变型p 5 3蛋白在不同胰腺疾病组织中的表达 ,探讨其生物学意义。方法应用标记的链霉卵白素 -生物素 (LSAB)免疫组化方法对 33例胰腺导管腺癌 (PC)、10例慢性胰腺炎 (CP)、9例正常胰腺组织进行p5 3及bc1- 2癌基因蛋白检测。结果bc1- 2和p 5 3蛋白阳性表达仅见于PC ,阳性率分别为 36 %和 5 5 % ;p 5 3蛋白表达与胰腺癌临床病理特征无关 ,而bc1- 2蛋白表达在组织学 1、2级和临床 1、2期显著高于组织学 3级和临床 3、4期。结论p 5 3和bc1- 2蛋白在部分胰腺癌的发生中可能起重要作用 ;bc1- 2蛋白表达可能为胰腺导管腺癌的早期表现。
OBJECTIVE: To investigate the biological significance of the expression of the apoptosis-inhibiting gene bc1-2 and the mutant p53 protein in different pancreatic diseases. Methods 33 cases of pancreatic ductal adenocarcinoma (PC), 10 cases of chronic pancreatitis (CP) and 9 cases of normal pancreatic tissue were subjected to immunohistochemistry with streptavidin-biotin (LSAB) for p5 3 and bc1-2 Oncoprotein detection. Results The positive rates of bc1-2 and p53 protein were only found in PC, the positive rates were 36% and 55% respectively. The expression of p53 protein was not related to the clinicopathological features of pancreatic carcinoma, while the expression of bcl- Grade 2 and clinical stage 1 and 2 were significantly higher than histological grade 3 and clinical stage 3 and 4. Conclusion p53 and bc1-2 proteins may play an important role in the development of some pancreatic cancers. The expression of bcl-2 protein may be an early manifestation of pancreatic ductal adenocarcinoma.
