目的探讨盐诱导高血压大鼠模型白细胞介素17A(IL-17A)、肿瘤坏死因子α(TNF-α)、转化生长因子β_1(TGF-β_1)和基质金属蛋白酶(MMP)9的表达以及IL-17A在该模型大鼠左心室间质胶原重构中的作用。方法将48只5周龄Wistar大鼠,随机分为正常对照组、2%盐浓度组和3%盐浓度组各16只,分别用蒸馏水、2%或3%NaCl饲养10周后,尾袖法测尾动脉血压,处死取左心室心肌组织,HE染色观察心肌细胞形态,Masson染色观察心肌胶原纤维并计算胶原容积分数(CVF),酶联免疫吸附试验(ELISA)、免疫组化法分别检测血浆和心肌组织IL-17A、TNF-α、MMP-9和TGF-β_1表达。结果2%盐浓度组高血压成模率75.00%(12/16),死亡率6.25%(1/16),3%盐浓度组成模率81.25%(13/16),死亡率12.50%(2/16);饲养10周后2%盐浓度组和3%盐浓度组大鼠的收缩压与对照组相比明显增高[(182.1±20.2)、(183.3±31.7)比(147.1±10.9)mm Hg,均P<0.01],但2%盐浓度组和3%盐浓度组两组间差异无统计学意义。2%盐浓度组和3%盐浓度组大鼠血浆和左心室心肌的IL-17A、TNF-α、MMP-9和TGF-β_1表达增高。IL-17A与CVF、MMP-9、TNF-α、TGF-β_1表达呈正相关(r=0.3、0.4、0.5、0.5,均P<0.05);CVF与TNF-α、TGF-β_1、MMP-9呈正相关(r=0.6、0.7、0.8;均P<0.01)。结论盐诱导高血压大鼠血浆和左心室心肌IL-17A表达明显增高,与TNF-α、MMP-9、TGF-β_1表达呈正相关。
Objective To investigate the expression of IL-17A, TNF-α, TGF-β_1 and MMP-9 in salt-induced hypertensive rats and the effect of IL -17A in the remodeling of left ventricular interstitial collagen in the model rats. Methods 48 Wistar rats of 5 weeks old were randomly divided into normal control group, 2% saline group and 3% saline group, 16 rats in each group. After feeding with distilled water, 2% or 3% NaCl for 10 weeks, The tail artery blood pressure was measured and the left ventricular myocardium was sacrificed. The morphology of myocardial cells was observed by HE staining. The collagen fibers of myocardium were observed by Masson staining, and the collagen volume fraction (CVF), enzyme-linked immunosorbent assay (ELISA) and immunohistochemistry Plasma and myocardial tissue IL-17A, TNF-α, MMP-9 and TGF-β 1 expression. Results The rate of hypertension was 75.00% (12/16), the mortality rate was 6.25% (1/16) in 2% salt concentration group, the rate of 81.25% (13/16) and the mortality rate of 12.50% / 16). Compared with the control group, systolic blood pressure of rats in 2% salt concentration group and 3% salt concentration group increased significantly ([(182.1 ± 20.2), (183.3 ± 31.7) vs (147.1 ± 10.9) mm Hg, all P <0.01]. However, there was no significant difference between the two groups in 2% salt concentration and 3% salt concentration groups. The levels of IL-17A, TNF-α, MMP-9 and TGF-β 1 in plasma and left ventricular myocardium were increased in 2% saline and 3% saline groups. The expression of IL-17A was positively correlated with the expression of CVF, MMP-9, TNF-α and TGF-β 1 (r = 0.3,0.4,0.5,0.5, all P < (R = 0.6,0.7,0.8; all P <0.01). Conclusion The expression of IL-17A in plasma and left ventricular myocardium of salt-induced hypertension rats was significantly increased, which was positively correlated with the expression of TNF-α, MMP-9 and TGF-β 1.
