目的:微量蛋白质对伊文思兰(EB)的共振散射光产生增强作用,且增强程度与蛋白质浓度有良好的线性关系,研究相应的光谱特征,探讨影响因素和适宜的反应条件。方法:建立了测定蛋白质的灵敏共振光散射(RLS)分析方法。结果:不同蛋白质在0 mg/L~10 mg/L范围内与散射强度呈直线关系。方法灵敏度较高,其检出限在54.1μg/L~105.8μg/L之间,线性范围宽,选择性和重现性较好,可用于多种蛋白质的测定。结论:利用灵敏共振光散射分析方法测定合成样品及人血清样品中的蛋白质,结果令人满意。
OBJECTIVE: Microprotein can enhance the resonance scattering light of Evans blue (EB) with a good linear relationship with the concentration of protein. The corresponding spectral characteristics are studied, and the influencing factors and appropriate reaction conditions are discussed. Methods: A sensitive resonance light scattering (RLS) method for the determination of proteins was established. Results: Different proteins showed a linear relationship with the scattering intensity in the range of 0 mg / L to 10 mg / L. The method has high sensitivity and the detection limit is between 54.1μg / L and 105.8μg / L. The method has wide linear range, good selectivity and reproducibility and can be used for the determination of various proteins. Conclusions: The results of sensitive resonance light scattering analysis of proteins in synthetic and human serum samples were satisfactory.
