目的:探讨红豆杉提取物对乳腺癌T47D细胞生长与凋亡的影响。方法:以乳腺癌T47D细胞为研究对象,实验设空白组、不同质量浓度红豆杉提取物组(200,100,50,25 mg·L-1)和阿霉素组(3μmol·L-1),各给药组处理乳腺癌T47D细胞24 h,MTT法检测T47D细胞增殖,试剂盒检测乳酸脱氢酶(LDH)及半胱天冬酶-3(Caspase-3)活性,Hoest33342荧光染色检测细胞凋亡,Western blot法检测T47D细胞凋亡相关蛋白Bax,Bcl-2,Caspase-3的表达。结果:与空白组比较,红豆杉提取物明显抑制乳腺癌T47D细胞的增殖,增加LDH及Caspase-3活性,诱导细胞凋亡及上调促凋亡蛋白Bax的水平,下调抗凋亡蛋白Bcl-2水平,均具有明显的统计学差异(P<0.05,P<0.01),且具有浓度依赖性。结论:红豆杉提取物对乳腺癌细胞T47D增殖具有直接抑制作用,并且具有浓度依赖性,其作用机制与促进细胞凋亡有关。
Objective: To investigate the effects of Taxus cuspidata extract on the growth and apoptosis of breast cancer T47D cells. Methods: Breast cancer T47D cells were used as experimental subjects. The blank group, different concentrations of Taxus cuspidata extract group (200,100,50,25 mg · L-1) and doxorubicin group (3μmol·L-1) The T47D cells of breast cancer were treated for 24 h in the treatment group, the proliferation of T47D cells was detected by MTT assay, the activity of lactate dehydrogenase (LDH) and caspase-3 were detected by kit, the apoptosis was detected by Hoest33342 staining Western blot was used to detect the expression of Bax, Bcl-2 and Caspase-3 in T47D cells. Results: Compared with the blank group, the extract of Taxus significantly inhibited the proliferation of breast cancer T47D cells, increased the activity of LDH and Caspase-3, induced apoptosis and up-regulated the level of pro-apoptotic protein Bax, down-regulated the expression of anti-apoptotic protein Bcl-2 (P <0.05, P <0.01), and with a concentration-dependent manner. Conclusion: Taxus extract has a direct inhibitory effect on the proliferation of breast cancer cell T47D cells in a concentration - dependent manner. Its mechanism of action is related to the promotion of apoptosis.
