用免疫组化和原位杂交方法观察了ＡＰＩ０１３４（ＡＰＩ）对凝血酶诱导猪主动脉内皮细胞表达原癌基因ｃｓｉｓｍＲＮＡ和血小板源生长因子（ＰＤＧＦ）Ｂ链蛋白的影响。结果表明，凝血酶能诱导内皮细胞ｃｓｉｓｍＲＮＡ转录和促进ＰＤＧＦＢ链蛋白表达，ＡＰＩ则能显著抑制凝血酶的上述诱导作用，剂量增大抑制作用增强。提示ＡＰＩ的这一作用可能是其抗动脉粥样硬化和预防血管成形术后再狭窄的机理之一 The effect of API0134 (API) on the expression of proto-oncogene c -sismRNA and platelet-derived growth factor (PDGF) -B chain protein in porcine aortic endothelial cells was investigated by immunohistochemistry and in situ hybridization. The results showed that thrombin can induce c -sismRNA endothelial cell transcription and promote the expression of PDGF B chain protein, API can significantly inhibit the above-mentioned induction of thrombin, increased dose inhibition. Suggesting that this effect of the API may be one of its mechanisms against anti-atherosclerosis and restenosis after angioplasty
