目的 研究丁酸类衍生物联合维甲酸对急性早幼粒细胞白血病 (APL )患者原代培养细胞的诱导分化作用。方法 采集 2 8例 APL患者骨髓单个核细胞体外培养 ,将各种诱导剂稀释后加入培养体系达到所需的效应浓度 ,台盼兰拒染率判断细胞存活率 ,离心甩片制片瑞氏染色后光镜下观察细胞形态 ,检测 NBT还原率及 CD11b评价细胞分化程度。结果 各组诱导剂对细胞存活力与对照组之间无明显差异 ,加入 ATRA、BA/ATRA、TB/ATRA组 ,细胞形态向终末期分化成熟、NBT还原率、CD11b表达升高 ,以 BA/ATRA、TB/ATRA组成熟细胞所占比例、NBT还原率高于单用 A-TRA组 ,而且 ATRA用量减少至单用时的 10 %。结论 丁酸类衍生物联合维甲酸对 APL 细胞具有协同诱导分化作用
Objective To study the effect of butyric acid derivatives and retinoic acid on the differentiation and differentiation of primary cultured human acute promyelocytic leukemia (APL) patients. Methods Bone marrow mononuclear cells from 28 APL patients were cultured in vitro. After dilution of various inducers into culture system, the desired effect concentration was achieved. The cell viability was determined by trypan blue exclusion rate. Under the light microscope, the cell morphology was observed, the NBT reduction rate and CD11b were measured to evaluate the degree of cell differentiation. Results There was no significant difference in cell viability between control group and ATRA, BA / ATRA and TB / ATRA groups. The cell morphology was differentiated to mature stage, NBT reduction rate and CD11b expression were increased, ATRA, TB / ATRA group of mature cells, NBT reduction rate was higher than the A-TRA alone group, and ATRA dosage reduced to 10% alone. Conclusion The butyric acid derivatives combined with retinoic acid have synergistic effect on inducing differentiation of APL cells
