目的快速检测啤酒花浸膏、溴酸钾和焦磷酸钠等3种食品添加剂对V79细胞DNA的损伤作用。方法采用V79细胞培养和单细胞凝胶电泳技术,借鉴单细胞凝胶电泳技术(SCGE)图像分析系统IMI1.0对彗星图像进行分析,观察指标有尾长、尾重心、olive尾矩、尾惯量、尾分布矩、尾长/头长比、尾DNA含量(%)等7个计量指标。结果实验结果表明,溴酸钾和焦磷酸钠的剂量分别在75~300μg/ml和25~100μmol/ml范围内,均对V79细胞DNA有明显的损伤作用,存在剂量-反应关系;啤酒花浸膏的剂量在25~100μg/ml范围内,不引起V79细胞DNA的损伤作用。结论本研究方法与传统的彗星实验相比,具有分析指标多,操作方便等优点,可应用于快速检测食品添加剂诱变性的筛选试验。
OBJECTIVE To rapidly detect DNA damage of V79 cells by using 3 kinds of food additives such as hop extract, potassium bromate and sodium pyrophosphate. Methods V79 cell culture and single cell gel electrophoresis were used to analyze comet image with IMI1.0, a single cell gel electrophoresis (SCGE) image analysis system. The indices of tail length, tail center of gravity, tail of tail, tail inertia , Tail distribution moment, tail length / head length ratio, tail DNA content (%) and other 7 indicators. Results The experimental results showed that the dose of potassium bromate and sodium pyrophosphate were significantly decreased in the range of 75 ~ 300μg / ml and 25 ~ 100μmol / ml, respectively, and the dose-response relationship existed. The dose of hops extract In 25 ~ 100μg / ml range, does not cause V79 cell DNA damage. Conclusion Compared with the traditional comet assay, this method has the advantages of more analytical indexes and more convenient operation. It can be used in the screening test to detect mutagenicity of food additives rapidly.
