目的:探讨脂肪肝大鼠瘦素水平的变化及丹参注射液的影响.方法:♂SD大鼠随机分为5组,其中1组为正常组,服用普通饲料,ip生理盐水;余4组用高脂饲料建立脂肪肝模型后,随机分为模型组、大剂量治疗组、小剂量治疗组、对照组,分别ip生理盐水、大、小剂量丹参注射液、古拉啶溶液.8wk后取血处死大鼠,测定血清瘦素等,观察脂肪肝病理变化.结果:脂肪肝模型大鼠肝指数较正常组升高(3.09%±0.24%vs 2.63%±0.19%,t=4.61,P =0.00),而血清瘦素水平较正常组低下(1.83±0.44μg/L vs 3.04±0.55μg/L,t=5.08,P= 0.00),大剂量丹参注射液提高模型组大鼠血清瘦素水平(2.29±0.46μg/L vs 1.83±0.44μg/ L,t=2.33,P=0.03).大剂量丹参治疗后脂肪肝大鼠肝组织脂肪沉者面积减少(1.45±0.93 vs 2.40±0.70,t=2.60,P=0.02).结论:丹参注射液改善脂肪代谢可能通过提高瘦素水平而实现.
Objective: To investigate the changes of leptin levels in fatty liver rats and the effects of Danshen injection. METHODS: Sprague-Dawley rats were randomly divided into 5 groups. One group was a normal group, which received normal diet and ip saline. The other 4 groups were randomly divided into a model group and a large-dose group after establishing a fatty liver model with high-fat diet. , low dose treatment group, control group, respectively ip saline, large and small doses of Danshen injection, Gu La Ding solution. After 8 weeks, the rats were killed by blood sampling and serum leptin was measured to observe the pathological changes of fatty liver. Results: Liver index of fatty liver model rats was higher than that of normal group (3.09%±0.24% vs 2.63%±0.19%, t=4.61, P=0.00), while serum Leptin levels were lower than those in the normal group (1.83±0.44 μg/L vs 3.04±0.55 μg/L, t=5.08, P=0.00). Large doses of Danshen Injection increased the model group size Rat serum leptin levels (2.29±0.46 μg/L vs 1.83±0.44 μg/L, t=2.33, P=0.03). After treatment with high-dose Salvia miltiorrhiza, the area of fatty liver in fatty liver rats decreased (1.45±0.93 vs 2.40±0.70, t=2.60, P=0.02). Conclusion: Salvia miltiorrhiza injection can improve fat metabolism by increasing leptin levels.
