目的 研究前胶原基因表达和胶原沉积与狼疮肾炎进展的关系。方法 狭线印迹杂交方法检测BXSB小鼠肾脏组织Ⅰ、Ⅲ型前胶原基N mRNA转录水平。斑点印迹法测定肾组织 Ⅰ、Ⅲ型胶原沉积情况。结果狼疮肾炎发病较重的 6月龄雄性 BXSB小鼠肾脏组织表达 Ⅰ、 Ⅲ型前胶原 mRNA的量显著增加( P均< 0. 05),分别为 3月龄雄鼠肾脏表达量的3.2倍和1.5倍,肾内Ⅰ、Ⅲ型胶原沉积较对照鼠显著增多(P分别<0.01)。结论Ⅰ、Ⅲ型前胶原mRNA的异位大量表达及Ⅰ、Ⅲ型胶原在肾脏沉积增加,是狼疮肾炎肾小球细胞外基质病理性积聚的重要原因,可能会引起肾小球结构和功能多方面的改变,在狼疮肾炎的进展和肾小球硬化的形成过程中起重要作用。
Objective To investigate the relationship between procollagen gene expression and collagen deposition and the progression of lupus nephritis. Methods The sIRS mRNA expressions of type Ⅰ and type Ⅲ procollagen were detected in the kidney of BXSB mice by slot blot hybridization. Dot blotting method was used to determine the type I and type Ⅲ collagen deposition in renal tissue. Results The expression of procollagen type Ⅰ and type Ⅲ procollagen mRNA in 6-month-old male BXSB mice with severe lupus nephritis incidence increased significantly (all P <0.05), which were respectively 3 in the 3-month-old male rats. 2-fold and 1.5-fold respectively. The deposition of type I and type III collagen in kidney increased significantly (P <0.01, respectively). Conclusion The ectopic expression of type Ⅰ and Ⅲ procollagen mRNA and the increase of deposition of type Ⅰ and type Ⅲ collagen in the kidneys are the important reasons for the pathological accumulation of glomerular extracellular matrix in lupus nephritis and may lead to glomerular structure and function more Changes in aspects of the progression of lupus nephritis and the formation of glomerular sclerosis play an important role in the process.
