目的 观察去甲基斑蝥素 (NCTD)体内外对人卵巢癌细胞株 3AO和 AO的抑制作用。方法 利用细胞培养技术 ,以结晶紫染色法测定 NCTD对 3AO和 AO细胞的体外杀伤作用 ,并在光镜和电镜下观察 NCTD对其进行治疗 ,以观察 NCTD体内对卵巢癌的抑癌效应。结果 体外实验表明 NCTD对 3AO和 AO细胞有较强抑制作用 ,并呈浓度和时间依赖性。在 NCTD作用下 ,光镜下卵巢癌细胞大多变圆呈悬浮状 ,电镜下则表现为细胞微绒毛消失 ,线粒体水肿、嵴断裂 ,胞质呈空泡样变性。体内实验则表明无论是腹腔内注射还是瘤内注射 ,NCTD对卵巢癌均有抑制瘤效应。结论 NCTD体内外对卵巢癌均有较强的抑制作用 ,提示 NCTD治疗卵巢癌可能有效
Objective To observe the inhibitory effect of NCTD on human ovarian cancer cell lines 3AO and AO in vitro and in vivo. Methods The cytotoxicity of NCTD to 3AO and AO cells was determined by crystal violet staining in vitro. The cells were treated with NCTD under light and electron microscope to observe the anti-cancer effects of NCTD on ovarian cancer. Results In vitro experiments showed that NCTD strongly inhibited 3AO and AO cells in a concentration- and time-dependent manner. Under the action of NCTD, most of the ovarian cancer cells were round and suspended in the light microscope, while the microvilli were disappeared under the electron microscope, mitochondria edema and cristae were broken, and cytoplasm was vacuolated. In vivo experiments showed that either intraperitoneal injection or intratumoral injection, NCTD on ovarian cancer inhibit tumor effect. Conclusions NCTD has a strong inhibitory effect on ovarian cancer both in vitro and in vivo, suggesting that NCTD may be effective in treating ovarian cancer
