目的研究夏枯草水提物对吡唑-脂多糖诱导的小鼠肝脏损伤的保护作用。方法将42只小鼠随机分为6组:正常对照组、模型组、夏枯草水提物低、中、高剂量组以及阳性对照组。灌胃给予小鼠夏枯草水提物,同时采用腹腔注射吡唑-脂多糖造成小鼠肝损伤模型,检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)和肝组织过氧化氢(H2O2)、丙二醛(MDA)、过氧化氢酶(CAT)、总超氧化物歧化酶(T-SOD)和谷胱甘肽(GSH)的活性/含量以及细胞色素酶CYP2E1和CYP2A5的蛋白表达水平。结果吡唑-脂多糖能够引起提高CYP2E1和CYP2A5的蛋白表达以及肝脏氧化应激并造成肝损伤;夏枯草水提物能够显著降低CYP2E1和CYP2A5的蛋白表达,降低氧化产物H2O2和MDA的含量,以及提高抗氧化系统中CAT、T-SOD活性和GSH含量。结论研究表明夏枯草可能通过抑制CYP2E1和CYP2A5的蛋白表达降低吡唑-脂多糖诱导肝组织内的氧化应激,从而对吡唑-脂多糖引起的肝损伤起保护作用。
Objective To study the protective effect of Prunella vulgaris aqueous extract on pyrazole - lipopolysaccharide - induced liver injury in mice. Methods 42 mice were randomly divided into 6 groups: normal control group, model group, Prunella vulgaris aqueous extract low, medium and high dose group and positive control group. The aqueous extract of Prunella vulgaris was given intragastrically and the hepatic injury model was induced by intraperitoneal injection of pyrazole-lipopolysaccharide. Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and hydrogen peroxide (H2O2) MDA, CAT, T-SOD and GSH, and the expressions of CYP2E1 and CYP2A5 Level. Results Pyrazole-lipopolysaccharide could increase the protein expression of CYP2E1 and CYP2A5 as well as hepatic oxidative stress and cause liver injury. Prunella vulgaris aqueous extract could significantly decrease the protein expression of CYP2E1 and CYP2A5 and reduce the content of H2O2 and MDA, Improve antioxidant system CAT, T-SOD activity and GSH content. Conclusions Prunella vulgaris could reduce the oxidative stress induced by pyrazole-lipopolysaccharide in liver tissue by inhibiting the protein expression of CYP2E1 and CYP2A5, and thus protect against hepatic injury induced by pyrazole-lipopolysaccharide.
