目的:研究补中益气汤对A549荷瘤小鼠脾组织中Fas蛋白表达的影响。方法:建立A549荷瘤小鼠模型进行体内抗肿瘤实验,采用随机对照法将30只BALB/c小鼠分为正常组、空白对照组、中药高剂量组、中药低剂量组、西药组,除正常组外,余组荷瘤,成功后,每天灌胃相应药物1次,连续10天后取材。采用免疫组织化学检测脾组织中Fas蛋白的表达;逆转录-多聚合酶链式反应(RT-PCR)法检测脾组织中Fas的mRNA表达。结果:正常组中Fas蛋白在细胞膜有表达,空白对照组出现较多棕黄色颗粒,与正常组比较,Fas表达明显升高(P<0.01);补中益气汤及顺铂作用后,Fas蛋白表达减少,与空白对照组比较,中药低剂量组、西药组Fas表达明显下降(P<0.05),中药高剂量组下降最显著(P<0.01)。中药高剂量组、中药低剂量组、西药组比较,Fas表达无明显变化(P>0.05)。与正常组比较,空白对照纽Fas的mRNA的表达量明显增加(P<0.01);经补中益气汤及顺铂作用后,Fas mRNA的表达减少,中药高剂量组、中药低剂量组及西药组与空白对照组的差异有统计学意义(P<0.01,P<0.05),而该3组的Fas mRNA的表达与正常组无明显差异。结论:补中益气汤可下调A549荷瘤小鼠脾脏中Fas蛋白的表达,从而促进机体免疫系统对肺癌细胞的杀伤。
Objective: To study the effect of Buzhong Yiqi Decoction on the expression of Fas protein in A549 tumor-bearing mice. Methods: A549 tumor-bearing mice model was established for in vivo anti-tumor experiments. Thirty BALB / c mice were randomly divided into normal group, blank control group, high dose Chinese medicine group, low dose Chinese medicine group and western medicine group The normal group, the remaining group of tumor-bearing, successful, gavage the corresponding drug 1 day for 10 days after drawing. The expression of Fas protein in spleen tissues was detected by immunohistochemistry. The expression of Fas mRNA in spleen tissues was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results: The expression of Fas protein in the normal group and the more brown granules in the blank control group were significantly higher than those in the normal group (P <0.01). The expression of Fas protein was up-regulated after Buzhong Yiqi Decoction and cisplatin treatment (P <0.05). Compared with the blank control group, the expression of Fas in low dose group and western medicine group decreased significantly (P <0.05), and the decrease in the high dose group was the most significant (P <0.01). There was no significant difference in the expression of Fas between TCM high dose group, TCM low dose group and western medicine group (P> 0.05). Compared with the normal group, the expression of Fas mRNA in blank control group was significantly increased (P <0.01). The expression of Fas mRNA was decreased after treated with Buzhong Yiqi Decoction and cisplatin. (P <0.01, P <0.05), while the expression of Fas mRNA in the three groups had no significant difference with the normal group. Conclusion: Buzhong Yiqi decoction can down-regulate the expression of Fas protein in spleen of A549 tumor-bearing mice and promote the killing of lung cancer cells by immune system.
