该文利用超临界CO2萃取装置对川西老鹳草中鞣质的提取工艺条件进行优化。采用磷钼钨酸-干酪素比色法测定鞣质的含量,以萃取压力、温度和时间为考察因素,以川西老鹳草萃取物中的鞣质含量为评价指标,通过正交试验对工艺条件进行优选。试验所得最佳工艺条件为萃取压力25 MPa,萃取温度50℃,萃取1.5 h。提取物中鞣质质量分数为12.91 mg·g-1,浸膏产率为3.67%。该比色法可以作为川西老鹳草鞣质的含量测定的方法,优化后的工艺稳定可行,提取率高,可为川西老鹳草中鞣质的提取工艺设计提供理论依据。
In this paper, using supercritical CO2 extraction device to optimize the extraction of tannin from Chuanxi geranium. The content of tannin was determined by phosphomolybdotungstic acid-casein colorimetric method. The extraction pressure, temperature and time were taken as the investigation factors. The content of tannin in the extract of Geranium chuanxi was used as the evaluation index. The orthogonal experiment Conditions are preferred. The best experimental conditions obtained for the extraction pressure 25 MPa, extraction temperature 50 ℃, extraction 1.5 h. The content of tannin in the extract was 12.91 mg · g-1, and the extract yield was 3.67%. The colorimetric method can be used as a method for determining the content of tannin in the geranium chuanxi, and the optimized process is stable and feasible with high extraction rate, which can provide a theoretical basis for the tannin extraction technology design of the geranium chuanxi.
