目的:观察42℃温热对人卵巢癌SKOV-3细胞热耐受和热耐受预测因子(HSP70)表达的影响。方法:(1)四氮唑蓝(MTT)快速比色法测细胞增殖率;(2)免疫细胞化学法测定HSP70的表达。结果:(1)42℃首次加热对人卵巢癌细胞SKOV-3有抑制和杀伤作用,短时间内(48h内)重复加热细胞,细胞会出现生长加快,以间隔8h重复加热组细胞生长加快尤为明显;(2)42℃加热SKOV-3细胞100min后48h内测HSP70表达,有不同程度的增加,以加热后8h细胞内HSP70的表达最强,有时间依赖性。结论:42℃加热100min可诱导人卵巢癌SKOV-3细胞内HSP70高表达,使细胞产生热耐受性,其出现与消退有时间依赖性。
Objective: To observe the effects of warming at 42 ℃ on the expression of heat tolerance and heat tolerance predictors (HSP70) in human ovarian cancer SKOV-3 cells. Methods: (1) Tetracycline blue (MTT) method was used to measure cell proliferation rate; (2) Immunocytochemical method was used to determine the expression of HSP70. Results: (1) The first heat treatment at 42 ℃ could inhibit and kill SKOV-3 in human ovarian cancer cells. In a short time (within 48h), the cells were heated repeatedly and the cells grew faster. The cells grew more rapidly at 8h intervals (2) HSP70 expression in SKOV-3 cells was detected within 48h after heating at 42 ℃ for 100min, with different degrees of increase, the expression of HSP70 was the strongest and time-dependent in 8h after heating. CONCLUSION: Heating at 42 ° C for 100 min induces high expression of HSP70 in human ovarian cancer SKOV-3 cells and leads to heat tolerance. The occurrence and extinction are time-dependent.
