报道了利用聚合酶链式反应（ＰＣＲ）鉴定重组ＴＰＯ阳性克隆的方法，同时比较了该方法与酶切鉴定的结果．取转化克隆小量提取质粒琼脂糖电泳分析，对怀疑含有重组子的工程菌进行ＰＣＲ鉴定，结果阳性克隆得到了特异性扩增片段．该方法与酶切鉴定结果完全一致，其优点是简便、灵敏、特异
A method for the identification of recombinant TPO positive clones by polymerase chain reaction (PCR) was reported. The results of the method and restriction enzyme digestion were also compared. A small amount of cloned plasmid was extracted and analyzed by agarose gel electrophoresis. PCR was carried out on the engineering bacteria suspected of containing the recombinants. As a result, the positive clones obtained the specific amplified fragments. The method is identical with the result of restriction enzyme digestion and has the advantages of simplicity, sensitivity and specificity
