目的研究DNA修复酶基因XRCC1、XRCC2、XRCC4、RAD52单核酸多态性在13-丁二烯(BD)致外周血淋巴细胞DNA损伤遗传易感性中的作用。方法选取171名BD作业工人和55名行政人员作为研究对象,采用碱性彗星试验评价个体外周血淋巴细胞DNA损伤,聚合酶链反应-限制性片段长度多态性方法分析XRCC1、XRCC2、XRCC4、RAD52基因多态性。结果 BD暴露组和对照组彗星尾距分别为4.53(3.49~5.87)和2.38(0.82~3.98),差异具有统计学意义(P<0.01)。暴露组中具有XRCC1基因G28152A位点GG基因型个体的彗星尾距显著低于AA基因型个体[4.52(3.43~5.64),5.18(4.23~6.18),P<0.05];XRCC4A245G位点AG和GG基因型个体的彗星尾距显著高于AA基因型个体[(5.05(3.79~6.05),5.37(4.29~6.36),4.23(3.35~5.41),P<0.01]。结论 XRCC1 G28152A和XRCC4 A245G位点的多态性可能与BD诱导的DNA损伤有关。
Objective To investigate the role of DNA repair enzyme genes XRCC1, XRCC2, XRCC4 and RAD52 in genetic susceptibility to DNA damage in peripheral blood lymphocytes induced by 13-butadiene (BD). Methods A total of 171 BD workers and 55 administrative staff were enrolled in this study. Alkaline comet assay was used to evaluate the DNA damage of peripheral blood lymphocytes and the polymorphisms of XRCC1, XRCC2, XRCC4, RAD52 gene polymorphism. Results The comet tail distances of BD exposure group and control group were 4.53 (3.49 ~ 5.87) and 2.38 (0.82 ~ 3.98) respectively, the difference was statistically significant (P <0.01). The comet tail distances of GG genotype individuals with G28152A locus in XRCC1 gene group were significantly lower than those in AA genotype individuals [4.52 (3.43-5.64), 5.18 (4.23-6.18), P <0.05]. The AGR and GG The tail distances of comets of genotype individuals were significantly higher than those of AA genotypes [(5.05 (3.79-6.05), 5.37 (4.29-6.36), 4.23 (3.35-5.41), P <0.01] .Conclusion The XRCC1 G28152A and XRCC4 A245G loci Polymorphism may be related to BD-induced DNA damage.
