目的探讨 1α (OH)D3对去卵巢( OVX)大鼠密质骨和松质骨的影响。方法将 41只雌性健康 Wistar大鼠分为 5组,于不同时间予以处死。( 1)基线组: 6月龄时处死;( 2)假手术组:施行手术,未切卵巢;( 3) OVXb组: 6月龄切除卵巢, 6周后处死;( 4) OVXe组:切除卵巢后 14周处死;( 5) O+ VD组 :于去卵巢 6周后经胃肠道给予 0.1μ g/(kg· d) 1α (OH)D3,共 8周。( 2)、( 4)和( 5)组均于手术后 14周时处死。对以上各组动物分别进行胫骨近端骨组织计量学、股骨 pQCT、骨生物力学测定和血尿骨转换指标的测定。结果去卵巢后大鼠的骨转换增加,骨小梁破坏,松质骨骨小梁减少,力学特性降低。经 1α (OH)D3治疗组血钙浓度有轻度升高,尿脱氧吡啶啉 /肌酐比率较未治疗组降低 67.0% [(43.50± 11.20)nmol· L- 1/mmol· L- 1vs(131.80± 14.90)nmol· L- 1/mmol· L- 1, P< 0.01],骨小梁的面积( 11.03± 0.73)比 OVXe组( 5.81± 1.29)增加 89.8%( P< 0.05)。股骨远端松质骨骨量和骨密度分别比 OVXe组增加 77.3%和 91.3%( P< 0.05)。 OVX和 1α (OH)D3均未引起密质骨骨量、面积和厚度的显著变化,而主要是表现为增龄性积累现象。 O+ VD组的股骨密质骨的极性截面惯性矩 (PMI)?
Objective To investigate the effects of 1α (OH) D3 on the densified bone and cancellous bone in ovariectomized rats. Methods Forty-one female healthy Wistar rats were divided into five groups and sacrificed at different times. (3) OVXb group: 6-month-old ovariectomized and sacrificed after 6 weeks; (4) OVXe group: resection of ovariectomized group (5) O + VD group: 0.1 μg / (kg · d) of 1α (OH) D3 was given via the gastrointestinal tract 6 weeks after ovariectomy for 8 weeks. Groups (2), (4) and (5) were sacrificed at 14 weeks after surgery. The above groups of animals were measured proximal tibia bone tissue, femur pQCT, bone biomechanical determination and conversion of hematuria and bone indicators. Results After ovariectomized rats, bone turnover increased, trabecular bone was destroyed, cancellous bone trabeculae were reduced, and mechanical properties were decreased. The serum calcium concentration increased slightly in the 1α (OH) D3 treatment group, and the urinary deoxypyridine / creatinine ratio decreased by 67.0% compared with the untreated group [(43.50 ± 11.20) nmol · L -1 / mmol · L -1 vs (131.80 ± 14.90) nmol · L -1 / mmol · L -1, P <0.01]. The area of trabecular bone (11.03 ± 0.73) was increased by 89.8% (P <0.05) compared with OVXe group (5.81 ± 1.29). The distal femur cancellous bone mass and bone mineral density increased by 77.3% and 91.3%, respectively, compared with OVXe group (P <0.05). OVX and 1α (OH) D3 did not cause significant changes in bone mass, area and thickness of the dense bone, but mainly manifested as age-related accumulation.
