目的探讨甲强龙对实验性变态反应性脑脊髓炎(EAE)小鼠的肿瘤坏死因子α(TNF-α)和白介素17(IL-17)的影响。方法将30只雌性C57BL/6J小鼠随机分为甲强龙组、EAE组和正常组,每组10只。EAE模型通过以髓鞘少突胶质细胞糖蛋白多肽35-55(MOG35-55)为抗原诱导。采用蛋白质印迹法(Western Blot)检测小鼠大脑的TNF-α和IL-17表达水平。结果甲强龙组的临床症状较EAE组改善(P<0.05),大脑的TNF-α和IL-17表达水平明显低于EAE组(P<0.05)。结论甲强龙能改善EAE小鼠的症状,其作用机制之一是抑制促炎性因子TNF-α和IL-17的表达。
Objective To investigate the effects of methylprednisolone on tumor necrosis factor α (TNF-α) and interleukin 17 (IL-17) in experimental allergic encephalomyelitis (EAE) mice. Methods Thirty female C57BL / 6J mice were randomly divided into methylprednisolone group, EAE group and normal group, with 10 rats in each group. The EAE model is induced by myelin oligodendrocyte glycoprotein 35-55 (MOG35-55) as an antigen. Western blot was used to detect the expression of TNF-α and IL-17 in mouse brain. Results The clinical symptoms of Jiaqianglong group were improved (P <0.05), and the levels of TNF-α and IL-17 in brain were lower than those of EAE group (P <0.05). Conclusions Methylprednisolone can ameliorate the symptoms of EAE mice. One of the mechanisms is that it inhibits the expression of pro-inflammatory cytokines TNF-α and IL-17.
