[目的]研究三种前体化合物对茅苍术组培苗中主要挥发油成分积累的影响,以期为人工栽培茅苍术的品质提高提供帮助。[方法]在茅苍术组培苗MS生根培养基中添加不同化合物,对组培苗进行一段时间培养后,超声提取植株所含的挥发油,利用气相色谱法测定挥发油中苍术酮、苍术醇、β-桉叶醇和苍术素的量。[结果]添加木糖、异戊二烯和四氢呋喃对茅苍术组培苗的生长指标、挥发油得率及四种组分相对百分含量均有影响。6g/L木糖组优化培养基中,检出挥发油中苍术酮、β-桉叶醇的相对百分含量分别达到4.23%和56.34%,分别比对照高出1.41和1.66个百分点;四氢呋喃组优化培养基中苍术酮、苍术醇和苍术素的相对百分含量均有提高,四氢呋喃浓度为0.07g/L时总挥发油含量较对照相比增加49.97%;添加异戊二烯到培养基中虽能提高苍术酮的相对百分含量,但对总挥发油含量的积累起到一定的抑制作用,较对照分别减少23.67%、31.06%和7.10%。[结论]添加木糖和四氢呋喃对茅苍术组培苗中主要挥发油的积累具有促进作用,而异戊二烯则具有抑制作用。
[Objective] The research aimed to study the effect of three precursor compounds on the accumulation of the main volatile oil components in the tissue culture seedlings of Atractylodes lancea, in order to provide the help for improving the quality of the cultivated Atractylodes. [Method] Different compounds were added to the MS rooting medium of the tissue culture plantlets of Atractylodes lancea. The tissue culture seedlings were cultured for a period of time. The volatile oil contained in the plant was extracted by ultrasonic. The contents of atractylodes, Eucalyptol and atractylodes. [Result] The growth index, volatile oil yield and relative percentage of four components of Atractylodes lancea were affected by adding xylose, isoprene and tetrahydrofuran. In 6g / L xylose group optimization medium, the relative percentage of atractylenol and β-eucalyptol in volatile oil were detected respectively, reaching 4.23% and 56.34% respectively, which were 1.41 and 1.66% higher than that of control group respectively; The relative percentages of atractyloside, atractylodes and atractylosin in the culture medium all increased, while the total volatile oil content increased by 49.97% compared with the control when the concentration of tetrahydrofuran was 0.07g / L; while the addition of isoprene to the culture medium could increase The relative percentage of atractyloside, however, inhibited the accumulation of total essential oil, which was 23.67%, 31.06% and 7.10% lower than that of the control respectively. [Conclusion] The addition of xylose and tetrahydrofuran promoted the accumulation of the main volatile oil in the tissue culture seedlings of Atractylodes macrocephala, while the isoprene had an inhibitory effect.
