目的了解5-氟脲嘧啶、丝裂霉素和表阿霉素分别及联合对人胃癌MGC-803细胞凋亡的诱导作用。方法用一定剂量的5-氟脲嘧啶。丝裂霉素和表阿霉素分别及三药联合与人胃癌细胞株MGC-803共同孵育12h后,应用光镜进行细胞形态学观察。细胞DNA经特异性荧光染色后,用流式细胞仪分析细胞周期变化。结果人胃癌MGC-803细胞经5-氧脲嘧啶、丝裂霉素和表阿霉素作用后,出现典型的细胞凋亡形态学改变,定量分析凋亡细胞发生率分别为10.2%、9.1%和9.7%,而三药联合作用后凋亡细胞的发生率为21.4%。结论5-氟脲嘧啶、丝裂霉素和表阿霉素可诱导人胃癌细胞林凋亡,三药联用可提高凋亡细胞的发生率。
Objective To investigate the induction of apoptosis in human gastric cancer MGC-803 cells induced by 5-fluorouracil, mitomycin and epirubicin, respectively. The method uses a dose of 5-fluorouracil. After mitomycin C and epirubicin were administered together with three drugs and human gastric cancer cell line MGC-803 for 12 hours, light microscope was used to observe cell morphology. Cellular DNA was stained with specific fluorescence, and cell cycle changes were analyzed by flow cytometry. Results After treated with 5-oxouracil, mitomycin and epirubicin in human gastric cancer MGC-803 cells, typical morphological changes of apoptosis were observed. The quantification of apoptotic cells was 10.2%. 9.1% and 9.7%, and the incidence of apoptotic cells was 21.4% after the combination of the three drugs. Conclusion 5-fluorouracil, mitomycin and epirubicin can induce apoptosis of human gastric cancer cell lines. The combination of three drugs can increase the incidence of apoptotic cells.
