目的:建立高效液相色谱法测定肋柱花四味汤散中盐酸小檗碱的含量测定方法。方法:色谱柱选择C18柱(Thermo Quent,5μm,250mm×406mm),流速为1.0mL/min;流动相选择磷酸二氢钾(乙腈),浓度为0.05mol/L,pH值通过磷酸控制在3,以30∶70配比;温度设置为25℃,控制检测液的长度保持在345nm。结果:小檗碱在0.039 14~0.391 4μg范围内有着良好的线性关系(r=0.999),RSD为2.6%,加样回收率为99.8%。结论:该方法简便易操作,可重复,测定结果准确。
Objective: To establish a HPLC method for determination of Berberine Hydrochloride in Ribiaohuasiwei Decoction. Method: Column C18 (Thermo Quent, 5μm, 250mm×406mm) was selected on the column and the flow rate was 1.0mL/min. Potassium dihydrogen phosphate (acetonitrile) was chosen as the mobile phase, the concentration was 0.05mol/L, and the pH value was controlled by phosphoric acid at 3%. , 30:70 ratio; temperature is set to 25 °C, control the length of the test solution is maintained at 345nm. RESULTS: Berberine had a good linearity in the range of 0.039 14 to 0.391 4 μg (r=0.999), RSD was 2.6%, and the recovery rate was 99.8%. Conclusion: This method is simple and easy to operate, repeatable and accurate.
