背景:c-fos基因为神经元细胞最常见的基因,其表达情况可作为受损神经细胞活性和反映性的标志物之一。目的:从分子水平探讨缺血性脑损伤大鼠海马c-fos基因表达情况和参麦注射液的保护作用。设计:随机对照实验。单位:泸州医学院组织学与胚胎学教研室。材料:实验于2002-01/03在泸州医学院组织学与胚胎学实验室完成。选择雄性Wistar大鼠30只,随机分为对照组、缺血组和治疗组,每组10只。方法:缺血组和治疗组大鼠夹闭双侧颈总动脉30min后再灌注1h,建立缺血性脑损伤模型;治疗组在缺血30min前腹腔注射参麦注射液2mL/kg(由红参、麦冬等中药组成);缺血组不给药;对照组做假手术,不夹闭颈总动脉,也不给药。取大鼠脑海马组织制备石蜡切片,每只大鼠取1张切片,每组共随机统计100个神经元胞核。根据神经元胞核着色情况观察各组大鼠海马神经元c-fos基因的表达(+++为着色深,呈深棕色。++为着色较深,呈棕色者。+为着色较浅,呈浅棕色者。-为未着色)。主要观察指标:各组大鼠海马神经元c-fos表达。结果:30只大鼠全部进入结果分析。缺血组大鼠海马内神经元c-fos基因的表达明显多于对照组犤+++:24,7个/视野,P<0.05犦,而治疗组明显少于缺血组犤+++:13,24个/视野,P<0.05犦。结论:参麦注射液可减少缺血性脑损伤大鼠海马神经元c-fos基因的表达,具有对缺血性脑损伤组织神经细胞的保护作用。
Background: The c-fos gene is the most common gene in neurons. The expression of c-fos gene can be used as one of the markers of the activity and reflectivity of damaged neurons. Objective: To investigate the c-fos gene expression in hippocampus of ischemic brain injury rats and the protective effect of Shenmai injection on the molecular level. Design: Randomized controlled trials. Unit: Department of Histology and Embryology, Luzhou Medical College. MATERIALS: The experiment was performed at the Histology and Embryology Laboratory of Luzhou Medical College from January 2002 to March 03. Thirty male Wistar rats were randomly divided into a control group, an ischemic group and a treatment group, 10 in each group. METHODS: Ischemic brain injury models were established after 30 minutes of bilateral common carotid arteries were occluded in ischemic and treated rats. The treatment group was intraperitoneally injected with Shenmai injection 2 mL/kg 30 min before ischemia. Ginseng, Ophiopogon japonicus and other traditional Chinese medicines); no ischemia in the ischemic group; sham operation in the control group, no clamping of the common carotid artery, nor dosing. Paraffin sections were prepared from rat hippocampus tissues. One rat was taken from each rat and 100 neurons were randomly counted in each group. The expression of c-fos gene in hippocampal neurons of each group was observed according to the staining condition of neurons. (+++ was dark and dark brown. ++ was darker and brown. + was lighter. Light brown. - Not colored. MAIN OUTCOME MEASURES: c-fos expression in hippocampal neurons of rats in each group. Results: All 30 rats entered the analysis of results. The expression of c-fos gene in neurons of hippocampus in the ischemic group was significantly higher than that of the control group (+++): 24, 7/field, P<0.05犦, but significantly lower in the treatment group than in the ischemic group. : 13, 24/field, P < 0.05 犦. Conclusion: Shenmai injection can reduce the expression of c-fos gene in hippocampal neurons of rats with ischemic brain injury, and it can protect nerve cells from ischemic brain damage.
