目的 研究邻苯二甲酸二(2- 乙基己基)酯 (DEHP)对果蝇寿命和体内脂质过氧化反应的影响。方法 经口喂饲果蝇 0 .20% 的DEHP,分别在染毒后 0、14、28d测定果蝇体内总超氧化物歧化酶(SOD)、CuZn SOD活性及其丙二醛含量,并同时进行果蝇生存实验。结果 DEHP可缩短果蝇寿命, 0 .05%、0 .10%和 0 .20% 3个染毒组雌雄果蝇半数死亡时间、平均寿命均低于对照组,并呈剂量 反应关系。对照组雌雄果蝇的半数死亡时间分别为 66d和 58d,实验组为 62~54d和 54~48d。雌雄果蝇的平均寿命也分别从对照组的 64d和 59d下降到 60~52d和 54~49d。0 .05%组雄性果蝇及 0. 10%、0 .20%组雌雄果蝇的平均最高寿命均低于对照组。DEHP还可抑制果蝇体内SOD活力并使丙二醛含量升高,呈剂量 反应关系。结论 DEHP可促进果蝇体内脂质过氧化反应,可能是缩短果蝇寿命、促进其衰老的原因之一。
Objective To study the effect of DEHP on the longevity and lipid peroxidation in Drosophila. Methods DEHP of 0.20% Drosophila was orally administrated, and the activity of superoxide dismutase (SOD) and CuZn SOD and the content of malondialdehyde (MDA) in Drosophila were determined at 0,14,28 d Drosophila survival experiment. Results DEHP could shorten the lifespan of Drosophila melanogaster. The half-life and average life span of Drosophila melanogaster at 0 .05%, 0 .10% and 0 .20% were lower than that of the control group and dose-response relationship. The control group of male and female flies half the time of death were 66d and 58d, the experimental group was 62 ~ 54d and 54 ~ 48d. The average lifespan of male and female flies also decreased from 64d and 59d respectively to 60 ~ 52d and 54 ~ 49d in the control group. 0 .05% of the male flies and 0. 10%, 0.20% of the male and female flies average maximum life expectancy were lower than the control group. DEHP also inhibited the vitality of SOD in Drosophila and the content of malondialdehyde increased in a dose-response relationship. Conclusion DEHP can promote lipid peroxidation in Drosophila, which may be one of the reasons for shortening the lifespan of Drosophila and promoting its aging.
