目的研究比较链霉亲合素(SA)-DTPA-Gd经腹腔和静脉注射后对荷瘤裸鼠体内肿瘤的强化效应,以提高磁共振(MR)靶向成像的敏感性。方法 12只荷人结肠癌裸鼠经尾静脉注射生物素化单抗600μg,24 h后腹腔注射80μg亲和素,30 min后分别经静脉和腹腔给予SA-DTPA-Gd。分别在给药前和给药后20、60 min和3、6、9、12、24 h进行MR 扫描并记录比较肿瘤和肝脏的信号强度。结果静脉组和腹腔组中肿瘤的最大强化率分别为70.2%和46.4%,两者具有显著性差异;静脉组和腹腔组中肝脏的最大强化率分别为20.4%和23.7%,两者间差异不显著。结论生物素化单克隆抗体和SA-DTPA-Gd对肿瘤具有特异性靶向增强作用;血药浓度较高是静脉注射SA-DTPA-Gd比腹腔注射更有利于皮下肿瘤强化的原因。
Objective To compare the enhancing effect of streptavidin (SA) -DTPA-Gd on tumors in nude mice after intraperitoneal and intravenous injection in order to improve the sensitivity of magnetic resonance (MR) targeted imaging. Methods Twelve human colon cancer nude mice were injected with 600μg of biotinylated monoclonal antibody via the tail vein and injected intraperitoneally with 80μg avidin 24 hours later. SA-DTPA-Gd was administered intravenously and intraperitoneally 30 minutes later. MR scanning was performed before administration and at 20, 60 min and 3, 6, 9, 12 and 24 h after administration, respectively, and the signal intensities of tumor and liver were compared. Results The maximum enhancement rates of tumors in the venous group and the abdominal cavity group were 70.2% and 46.4%, respectively, with significant differences between the two groups. The maximum enhancement rates of the liver in the venous group and the abdominal cavity group were 20.4% and 23% .7%, the difference between the two is not significant. Conclusions Biotinylated monoclonal antibody and SA-DTPA-Gd have specific target-enhancing effect on tumor. Higher plasma concentration is the reason that SA-DTPA-Gd intravenously is more beneficial to the enhancement of subcutaneous tumor than intraperitoneal injection.
