目的 :探讨去氢表雄酮 (DHEA)的体内抗癌作用及其机制。方法 :Buffalo大鼠 2 1只随机分为空白对照组 (n=5 ) ;肿瘤对照组 (把 Morris肝细胞瘤移植到大鼠的两肋皮下 ,建立 Morris肝细胞瘤移植的 Buffalo大鼠的体内模型 ) ,喂养基础饲料 (n=6 )、DHEA肿瘤组 (建立 Morris肝细胞瘤移植模型后 ,在喂养的基础饲料中添加 DHEA) (n=6 )和去氢表雄酮硫酸酯 (DHEA- s)肿瘤组 (建立 Morris肝细胞瘤移植模型后 ,在喂养的基础饲料中添加 DHEA - s) (n=4 )。分别喂养 4周后 ,应用流式细胞术检测大鼠的免疫功能 ,应用磁场型高分解能质量分析仪 (GC/ MS)方法测定大鼠体内类固醇的含量 ,采用免疫组化方法染色检测磷酸化的磷酸酶和张力蛋白同源物基因 (PTEN )蛋白表达 ,同时测定其对大鼠的毒副作用。结果 :DHEA肿瘤组的瘤重量 [(8.31± 0 .6 1) g]较肿瘤对照组 [(14 .5 7± 0 .5 6 ) g]显著减小 ,抑制率达到 4 3%。免疫组织化学染色显示 DHEA肿瘤组的磷酸化PTEN蛋白表达量增加 ,同时提高了 CD3、CD4阳性细胞百分率及 CD4 + / CD8+比值 ,与肿瘤对照组相比差异有显著性 (P<0 .0 5 )。 DHEA肿瘤组的胆固醇、甘油三脂及高密度脂蛋白的含量与空白对照组相比差异无显著性 ,未见其有毒副作用。结论 :DHEA对肿瘤具有明显的抑制作?
Objective: To investigate the anti-cancer effect and mechanism of dehydroepiandrosterone (DHEA) in vivo. METHODS: Twenty two Buffalo rats were randomly divided into blank control group (n = 5), tumor control group (Morris hepatoma cells were transplanted subcutaneously in the two flanks of rats to establish Morris hepatoma transplanted Buffalo rats (N = 6), DHEA tumor (DHEA-fed basal diet after establishing Morris hepatoma model) and DHEA- s) tumor group (DHEA - s was added to the fed basal diet after establishment of the Morris hepatoma cell transplant model) (n = 4). After four weeks of feeding respectively, the immune function of the rats was detected by flow cytometry. The levels of steroid in rats were determined by magnetic field-type high-resolution mass spectrometry (GC / MS) method. The phosphorylated Phosphatase and tension protein homologue (PTEN) protein expression, while the determination of its toxic side effects on rats. Results: The tumor weight of DHEA tumor group [(8.31 ± 0.61) g] was significantly reduced compared with that of tumor control group [(14.57 ± 0.56) g] and the inhibition rate was 43%. Immunohistochemical staining showed that the expression of phosphorylated PTEN protein in DHEA tumor group increased, while the percentage of CD3, CD4 positive cells and the ratio of CD4 + / CD8 + increased significantly compared with the tumor control group (P <0.05) ). The content of cholesterol, triglyceride and high density lipoprotein in DHEA tumor group had no significant difference compared with the blank control group, and no toxic and side effects were found. Conclusion: DHEA has significant inhibition of the tumor?
