】目的 :以平阳霉素诱导食管癌细胞凋亡 ,并探索药物浓度与诱导凋亡之间的关系。材料与方法 :应用电镜、DNA电泳、TUNEL、荧光显微镜、激光共聚焦扫描显微镜等手段 ,观察食管癌细胞在不同浓度平阳霉素作用不同时间后的变化特征及诱导凋亡之作用。结果 :细胞形态学及DNA水平的变化均证实靶细胞发生了凋亡 ,但较大浓度较长时间的平阳霉素作用可致其坏死 ;平阳霉素对EC8712凋亡的诱导效率呈浓度、时间依赖性增强。结论 :一定浓度平阳霉素作用一定的时间可以诱导食管癌细胞株EC8712发生凋亡。诱导癌细胞凋亡可能是平阳霉素化疗作用的重要机制。临床化疗时可根据量效关系合理用药以诱导肿瘤细胞达到最大的凋亡率 ,而又不致其坏死、发生毒副作用
[Objective] To induce apoptosis of esophageal carcinoma cells by pingyangmycin, and to explore the relationship between drug concentration and induction of apoptosis. Materials and Methods : Electron microscopy, DNA electrophoresis, TUNEL, fluorescence microscopy, laser confocal scanning microscopy and other means were used to observe the changes of esophageal cancer cells at different concentrations of Pingyangmycin and their induction of apoptosis. RESULTS: Changes in cell morphology and DNA levels all confirmed that apoptosis occurred in the target cells, but the effect of Pingyangmycin at a relatively high concentration for a long time could cause necrosis; the induction efficiency of Pingyangmycin on EC8712 apoptosis was concentration and time. Dependency enhancement. Conclusion: A certain concentration of bleomycin can induce apoptosis of esophageal cancer cell line EC8712 for a certain period of time. Induction of cancer cell apoptosis may be an important mechanism of the role of Pingyangmycin chemotherapy. In clinical chemotherapy, reasonable doses can be used according to the dose-effect relationship to induce tumor cells to achieve the maximum rate of apoptosis without necrosis and toxic side effects.
