目的观察左、右归丸及其拆方对去卵巢大鼠肾脏PPARγ、FABP4基因与蛋白表达的影响,并探讨补肾法对绝经后妇女脂代谢紊乱的改善。方法将90只SD雌性大鼠随机分为空白组、假手术组、模型组、补佳乐组、左归丸组、右归丸组、共同药组、滋阴药组、补阳药组。除假手术、空白组外均进行双侧卵巢切除。经12周治疗分别以Q-PCR法、Western blot法检测PPARγ、FABP4在肾脏中基因和蛋白的表达。结果与空白组相比,除假手术组外,PPARγ、FABP4 mRNA及蛋白表达升高(P<0.01);与模型组比较,各用药组PPARγ、FABP4 mRNA及蛋白表达降低(P<0.01);与补佳乐组比较,各用药组PPARγ、FABP4表达高,其中左归丸组表达较低。结论左、右归丸及其拆方可以明显影响肾脏中脂代谢相关指标,其中以左归丸改变最为显著。
Objective To observe the effects of Zuogui Gui Wan and its decomposed formulas on the expression of PPARγ and FABP4 gene and protein in the kidney of ovariectomized rats and to explore the effect of tonifying kidney on the disturbance of lipid metabolism in postmenopausal women. Methods Ninety SD female rats were randomly divided into blank group, sham operation group, model group, Bu Jiale group, Zuoguiwan group, Youguiwan group, common medicine group, Ziyin drug group and Buyangyao group. In addition to sham surgery, both sides of the blank group were ovariectomized. After 12 weeks treatment, the expression of PPARγ and FABP4 gene and protein in kidney were detected by Q-PCR and Western blot respectively. Results Compared with the blank group, the mRNA and protein expressions of PPARγ and FABP4 increased (P <0.01) except the sham operation group. Compared with the model group, the expression of PPARγ and FABP4 mRNA and protein decreased (P <0.01). Compared with nocturnal group, each treatment group PPARγ, FABP4 expression was high, which Zuogui pill group was lower expression. Conclusion Zuogui Gui Wan and its disassembled formulas can obviously affect the index of lipid metabolism in the kidney, of which Zuo Gui Wan is the most significant change.
