目的研究β1,4-半乳糖基转移酶-Ⅰ(β1,4-GalT-Ⅰ)在类风湿性关节炎(RA)患者膝关节滑膜组织中表达水平及细胞定位。方法选择12例RA患者(A组)和8例健康人(B组)滑膜组织,采用Western blot检测β1,4-GalT-Ⅰ、E-选择素的蛋白表达水平,免疫组化检测β1,4-GalT-Ⅰ的表达和分布情况,免疫荧光检测β1,4-GalT-Ⅰ与滑膜组织中炎症细胞以及E-选择素的共定位情况。结果与B组相比,A组滑膜组织中β1,4-GalT-Ⅰ和E-选择素蛋白表达明显增加(P<0.01或P<0.05)。在RA滑膜炎症过程中,β1,4-GalT-Ⅰ主要表达于巨噬样滑膜细胞、成纤维样滑膜细胞、活性T细胞和中性粒细胞。E-选择素与β1,4-GalT-Ⅰ存在共定位。结论β1,4-GalT-Ⅰ可能参与调节RA滑膜组织的炎症反应过程。
Objective To investigate the expression and cellular localization of β1,4-galactosyltransferase-Ⅰ (β1,4-GalT-Ⅰ) in synovial tissue of patients with rheumatoid arthritis (RA). Methods Twelve synovial tissues of RA patients (group A) and eight healthy people (group B) were selected to detect the protein expression levels of β1,4-GalT-Ⅰ and E-selectin by Western blot. The expressions of β1, 4-GalT-Ⅰ expression and distribution, immunofluorescence detection of β1,4-GalT-Ⅰ and synovial tissue inflammatory cells and E-selectin co-localization of the situation. Results Compared with group B, the expression of β1,4-GalT-Ⅰ and E-selectin in group A was significantly increased (P <0.01 or P <0.05). During RA synovial inflammation, β1,4-GalT-I is mainly expressed in macrophage-like synoviocytes, fibroblast-like synoviocytes, reactive T cells and neutrophils. E-selectin co-localized with β1,4-GalT-I. Conclusion β1,4-GalT-Ⅰ may be involved in the regulation of inflammatory reaction in synovial tissue of RA.
