割手密是甘蔗近缘野生种之一,在甘蔗育种中具有重要地位和作用。本研究采用Genomic-SSR和EST-SSR两种类型分子标记对来自广东各个地区的67个割手密种质进行分析。在20对SSR引物上共检测到341个多态性条带,每个引物上的平均多态性为17.05,其中11个Genomic-SSR的平均多态性为20.82,9个EST-SSR的平均多态性为12.44。广东割手密在Genomic-SSR上的多态性显著高于EST-SSR上的多态性。MantelTest分析表明:通过Genomic-SSR、EST-SSR以及Genomic-SSR+EST-SSR三种方法所计算的试验材料的遗传距离矩阵之间都呈极显著相关(p<0.01),说明应用两种类型SSR获得的各个材料间的遗传关系具有较高的一致性。广东割手密在20对SSR引物上的遗传多样性指数介于0.1064~0.2916之间,平均多样性指数为0.1943;各个材料间的遗传距离为0.0063~0.3082之间,平均为0.1935。UPGMA聚类分析表明:广东割手密之间的遗传关系和地理来源存在一定的相关,由南向北存在较为显著的地理分化。
Cut the secret is one of the sugar cane relatives of wild species, in sugarcane breeding has an important position and role. In this study, Genomic-SSR and EST-SSR markers were used to analyze the genetic diversity of 67 cutwort germplasms from all regions of Guangdong. A total of 341 polymorphic bands were detected on 20 pairs of SSR primers. The average polymorphism on each primer was 17.05. The average polymorphism of 11 Genomic-SSRs was 20.82. The average of 9 EST-SSRs Polymorphism was 12.44. Polymorphism of Genome-SSR was significantly higher in Guangdong than in EST-SSR. MantelTest analysis showed that the genetic distance matrices calculated by Genomic-SSR, EST-SSR and Genomic-SSR + EST-SSR were significantly correlated (p <0.01), indicating that two types The genetic relationships among the various materials obtained by SSR are highly consistent. The genetic diversity index of 20 pairs of SSR primers was between 0.1064 and 0.2916 with the average diversity index of 0.1943. The genetic distance among all the materials was between 0.0063 and 0.3082 with an average of 0.1935. UPGMA cluster analysis showed that there was a certain correlation between genetic relationship and geographical origin of Guangdong’s cuttlebone, with significant geographical differentiation from south to north.
