目的 :研究三氧化二砷 (As2 O3)对人鼻咽低分化鳞癌可移植瘤在SCID小鼠体内生长的抑制作用 ,并初步研究其对细胞分化的影响。方法 :以人鼻咽低分化鳞癌细胞株CSNE 1为研究对象 ,观察腹腔注射As2 O3(5mg kg)对人鼻咽癌在SCID小鼠体内生长情况。用光学显微镜和电子显微镜观察形态结构变化、免疫组化方法检测增殖细胞核抗原 (PCNA)的表达情况。结果 :腹腔注射As2 O3后 ,鼻咽癌SCID小鼠移植瘤生长抑制 ,抑瘤率 75 4 %。同时 ,瘤组织中癌细胞密度减少 ,细胞皱缩 ,胞浆红染 ,瘤组织分化渐成熟 ,出现角化细胞和角化珠 ;间质结缔组织增多。透射电镜下肿瘤细胞均出现成熟分化 ,核浆比例减少 ,胞浆中细胞器增多 ,出现大量张力原纤维并围绕核周 ,可见细胞明显角质化 ,细胞表面微小突起增多 ,桥粒增多 ,细胞之间以桥粒互相连接。癌细胞PCNA在阴性对照组呈高表达 ,阳性细胞指数 (PI)达 95 % ,As2 O3组的细胞PC NA表达明显减少 ,PI5 5 % (P =0 .0 0 1)。结论 :As2 O3抑制SCID小鼠体内人鼻咽低分化鳞癌的生长 ,这种抑制作用可能与其能够诱导癌细胞向成熟方向分化有关
OBJECTIVE: To study the inhibitory effect of As 2 O 3 on the growth of human nasopharyngeal carcinoma with poorly differentiated squamous cell carcinoma in vivo in SCID mice and to study its effect on cell differentiation. Methods: Human nasopharyngeal poorly differentiated squamous cell carcinoma cell line CSNE 1 was used as the research object to observe the growth of human nasopharyngeal carcinoma in SCID mice by intraperitoneal injection of As 2 O 3 (5 mg kg). Morphological changes were observed with optical microscope and electron microscope. The expression of proliferating cell nuclear antigen (PCNA) was detected by immunohistochemistry. Results: As2 O3 was injected intraperitoneally. The growth of the transplanted tumor in SCID mice was inhibited and the tumor inhibition rate was 75.4%. At the same time, the tumor tissue density of cancer cells decreased, cell shrinkage, cytoplasmic red staining, tumor tissue differentiation gradually mature keratinocytes and keratinocytes; interstitial connective tissue increased. Tumor cells under the TEM showed mature differentiation, reduced proportion of nuclear plasma, increased organelles in the cytoplasm, a large number of fibril fibers around the perinuclear weeks, significant keratinization of cells, increased microprojections on the cell surface, increased desmosomes, Bridged to each other. PCNA in cancer cells was highly expressed in the negative control group with a positive cell index (PI) of 95%. The expression of PCNA in As2O3 group was significantly decreased, and the percentage of PI5 was 5% (P = 0.010). CONCLUSIONS: As2 O3 inhibits the growth of human nasopharyngeal poorly differentiated squamous cell carcinoma in vivo in SCID mice.
